新疆地区汉族帕金森病患者基因组长链非编码RNA的差异表达谱筛查分析

摘要 目的分析帕金森病患者外周血细胞中长链非编码RNA(lncRNA)的表达情况,根据lncRNA的差异表达并结合mRNA表达数据预测其可能的功能。方法收集中国新疆地区汉族帕金森病患者55例(帕金森病组)和健康志愿者55例(对照组)的外周血提取总RNA。用微阵列法测定10个样本(帕金森病组5例和对照组5例)中全基因组lncRNA和mRNA的表达水平。采用qRT-PCR检测100例(帕金森病组50例,对照组50例)lncRNA的表达水平。依据lncRNA微阵列芯片分析结果,建立帕金森病的lncRNA表达谱,筛选出具有差异表达的lncRNAs,应用临床大样本数据验证其结果与芯片数据结果的一致性。结果两组共鉴定出171个上调和120个下调的lncRNA,组间表达差异有统计学意义。共同表达的mRNA注释结果显示,GO分析中"生物过程调控"为一类显著相关的基因产品属性,炎症免疫反应是最显著的相关通路。结论与对照组比较,帕金森病组外周血中lncRNA的表达明显不同,差异表达的lncRNA可能在帕金森病的发生发展中发挥潜在作用。 To analyze the expression of long non-coding RNA(lncRNA) in peripheral blood cells of Parkinson’s disease(PD) patients, and predict its possible function based on the differential expression of lncRNA and combination with mRNA expression data. Methods Total RNA was extracted from the peripheral blood of 55 PD patients and 55 healthy volunteers(control group) of Han nationality in Xinjiang. The expression levels of genome-wide scale lncRNA and mRNA were determined by microarray in 10 samples(5 PD and 5 controls). qRT-PCR was used to validate lncRNA expression levels in 100 samples(50 PD and 50 controls). Results A total of 171 up-and 120 downregulated lncRNA were identified to be significantly and differentially expressed between the two groups. The annotation result of their co-expressed mRNA showed that the most significantly related category of GO analysis was regulation of biological processes, and the most significantly related pathway was inflammation and immune reaction. Conclusion Our study indicated that clusters of lncRNA were significantly and differentially expressed in the peripheral blood of PD patients. These newly identified lncRNA may have a potential role in the development of PD.
