摘要
目的探讨长链非编码RNA MCM3AP-AS1(MCM3AP antisense RNA 1,MCM3AP-AS1)靶向调控微小RNA-876-5p(miR-876-5p)对卵巢癌SKOV3细胞增殖和转移的影响。方法选取2017年4月至2018年5月温州市中心医院妇产科收治的40例卵巢癌患者癌组织和相应癌旁组织标本,及卵巢癌细胞系(CaOV3、SKOV3、OVCAR3、OV90)和正常卵巢上皮细胞系(HOSE)。采用实时定量聚合酶链式反应法(qRT-PCR)检测卵巢癌组织和细胞中lncRNA MCM3AP-AS1的表达水平;建立lncRNA MCM3AP-AS1低表达模型,采用MTT实验检测卵巢癌细胞增殖,采用transwell检测卵巢癌细胞的迁移和侵袭水平;采用生物信息分析、荧光素酶实验、qRT-PCR验证lncRNA MCM3AP-AS1和miR-876-5p的靶向关系。结果lncRNA MCM3AP-AS1在卵巢癌组织中的水平为(8.45±0.86),高于癌旁组织(4.45±0.45);在卵巢癌细胞中的水平为CaOV3(1.53±0.12),SKOV3(1.82±0.23),OVCAR3(1.34±0.12),高于正常卵巢上皮细胞HOSE(1.00±0.10);下调lncRNA MCM3AP-AS1能降低卵巢癌细胞增殖、迁移和侵袭;lncRNA MCM3AP-AS1能与miR-876-5p相互作用并下调其表达。结论lncRNA MCM3AP-AS1可通过靶向抑制miR-876-5p促进卵巢癌细胞的增殖、迁移和侵袭。
Objective To investigate the effects of long-chain non-coding RNA MCM3AP-AS1(MCM3AP antisense RNA 1,MCM3AP-AS1)targeting regulation of microRNA-876-5p(microRNA-876-5p)on the proliferation and metastasis of ovarian cancer cells.Methods Forty patients with ovarian cancer were selected from Apr.2017 to May.2018.The ovarian cancer cell lines(CaOV3,SKOV3,OVCAR3,OV90)and normal ovarian epithelial cell lines(HOSE)were also selected.Real-time quantitative polymerase chain reaction(qRT-PCR)was used to detect the expression of lncRNA MCM3AP-AS1 in ovarian cancer tissues and cells.A low expression model of lncRNA MCM3AP-AS1 was established.Proliferation and metastasis of ovarian cancer cells were detected by MTT and transwell.Luciferase assay and qRT-PCR were used to verify the targeting relationship between lncRNA MCM3AP-AS1 and miR-876-5p.Results The level of lnRNA MCM3AP-AS1 in ovarian cancer tissue was(8.45±0.86),higher than that in adjacent tissue(4.45±0.45);The levels in ovarian cancer cells were CaOV3(1.53±0.12),SKOV3(1.82±0.23),OVCAR3(1.34±0.12),higher than those in normal ovarian epithelial cell HOSE(1.00±0.10);Down-regulated lncRNA MCM3AP-AS1 could reduce the proliferation and metastasis of ovarian cancer cells;lncRNA MCM3AP-AS1 could interact with and down-regulate the expression of miR-876-5p.Conclusion LncRNA MCM3AP-AS1 can promote the proliferation,metastasis and invasion of ovarian cancer cells by targeting the inhibition of microRNA-876-5p.
