摘要
本研究旨在筛选出能够在较高铜离子浓度下生长并吸附铜离子的菌株,并对其吸附特性进行研究,以期为微生物处理重金属污染废水提供可靠的技术支持。本实验充分利用环境对微生物的胁迫作用,选择被铜离子污染的土壤,放入锥形瓶中并用水稀释后取上清夜涂布于含铜离子的4种培养基(即细菌、霉菌、放线菌和真菌固体培养基)。经过划线分离得到单菌落,从中挑选耐受能力最强的菌株,对其进行菌种16SrDNA鉴定及生长、吸附条件的优化。结果筛选到一株对铜离子耐受达到19 000mg/L的菌株,经鉴定为枯草芽孢杆菌,优化吸附条件后其对溶液中铜离子吸附率可以达到72%。实验筛选到的菌株具有改善和治理铜离子污染废水的潜力。
A strain that could survive and adsorb copper ions in the environment with high concentration of copper ions was selected and the adsorption characteristics of the strain were studied,so as to provide reliable technical support for microbial treatment of heavy metal polluted wastewater.This experiment fully utilized the environmental stress on microorganism.The soil contaminated with copper ions was chosen,then it was put it into an Erlenmeyer flask and diluted with water.The supernatant was taken and applied to 4 kinds of culture medium containing copper ions(bacteria,molds,actinomycetes and fungi solid medium).A single colony was obtained by streaking.The strains with the strongest resistance to copper ions were selected.The strain 16 SrDNA was identified and the growth and adsorption conditions were optimized.A strain with a copper ion tolerance of 19 000 mg/L was selected and identified as Bacillussubtilis.After optimizing the adsorption conditions,the adsorption rate of copper ion in the solution could reach 72%.The experimentally screened strains have the potential to improve and treat copper ion-contaminated wastewater.
