HPLC结合红外光谱法评价不同虎耳草质量

HPLC Analysis Combined with Infrared Atlas for Quality Assessment of Saxifraga Stolonifera

目的:利用HPLC同时测定虎耳草中绿原酸、槲皮素的含量,结合红外图谱分析,对西藏地区不同来源的虎耳草药材的质量进行比较。方法:采用超声辅助提取,HPLC结合红外光谱法测定,色谱柱为Agilent SB-C18(250 mm×4.6 mm,5μm)分析柱,以甲醇(A)∶0.1%磷酸溶液(B)为流动相,梯度洗脱,检测波长为350 nm,流速为1 mL/min,柱温为30℃。结果:利用HPLC法结合红外图谱比较分析7批不同来源的虎耳草药材发现,8月采收的篦齿虎耳草中绿原酸和槲皮素含量最高,藏中虎耳草的绿原酸和槲皮素含量最低。不同虎耳草红外吸收峰波数在4000~400 cm^-1存在差异。结论:HPLC法结合红外图谱分析可以初步评价虎耳草质量。

Objective:To determine the chlorogenic acid and quercetin in Saxifraga by HPLC combined with infrared spectroscopy to compare and evaluate the quality of Saxifraga from different sources.Methods:Ultrasound assisted extraction was used,and HPLC analysis was performed on a Aglen(250 mm×4.6 mm,5μm)column.The mobile phase was 1%Phosphoric acid and Methanol.A gradient elution was used.The detection wavelength was 350 nm.The flow rate is 1 mL/min and the sample injection rate was 10μL.Results:Totally 7 different sources of Saxifraga were measured by HPLC method combined with infrared spectroscopy.There is a great difference in chlorogenic acid and quercetin content,which were higher in the saxifrage umbellulata and lower in the Saxifraga signatella.The different infrared spectroscopy wave were exsited during 4000-400 cm^-1.Conclusion:This method can be used to evaluate the quality of Saxifraga.
