摘要
目的找到高脂血症与非高脂血症人群间的肠道菌群差异。方法采集中央民族大学校医院参加慢性病体检和大肠癌筛查的教师职工临床病例样本,高脂血症组80例,非高脂血症组44例。基于16S rDNA高通量测序技术(Illumina技术),运用SPSS 25.0的倾向性评分匹配(PSM)法对高脂血症患者与非高脂血症人群的样本临床资料匹配,并且进行相关数据处理及分析,探讨高脂血症组与非高脂血症组间的肠道菌群的差异。结果高脂血症组与非高脂血症组的差异分类单元(OTU)分属于4个门和30个属。根据贝叶斯网络推理算法,得到菌群间与高脂血症相互作用贝叶斯网络推理图。其中OTU630和OTU15与高脂血症联系最紧密且均为拟杆菌属,OTU15为高脂血症的直接致病菌。结论拟杆菌属为高脂血症与非高脂血症间的关键菌群,未来可以对该菌种做进一步深入研究,以达到预防和治疗高脂血症的目的。
Objective To identify the difference in intestinal flora between hyperlipidemia and non-hyperlipidemia populations.Methods A sampling of clinical specimens was conducted from teachers and staff members of the Minzu University Hospital who participated in the physical checkup for chronic diseases and colorectal cancer screening,including 80 subjects in the hyperlipidemia group and 44 in the non-hyperlipidemia group.Based on 16S rDNA high-throughput sequencing techniques(Illumina),the PSM module of SPSS 25.0 package was used to match the clinical data of hyperlipidemia patients with non-hyperlipidemia subjects who offered the samples,and process the related data for difference in intestinal flora between the hyperlipidemia and non-hyperlipidemia groups.Results The differences in operational taxonomic units(OTUs)between the hyperlipidemia and the non-hyperlipidemia groups belonged to 4 phyla and 30 genera.Based on the Bayesian network inference algorithm,a Bayesian network inference map of the interaction between flora and hyperlipidemia was obtained.Among them,OTU630 and OTU15 were most closely related to hyperlipidemia and the both were Bacteroides.Finally,OTU15 was identified as a direct pathogen of hyperlipidemia.Conclusion Bacteroides are key flora that distinguishes hyperlipidemia and non-hyperlipidemia.Future research on this species are worthwhile for the purpose of preventing and treating hyperlipidemia.
