宫颈癌组织STING、CCL22、IDO表达变化及其临床意义 被引量:7

摘要 目的观察宫颈癌组织干扰素基因刺激因子(STING)、CC类趋化因子22(CCL22)、吲哚胺-2,3-双加氧酶(IDO)表达变化,并探讨其临床意义。方法选择宫颈癌患者83例,取手术切除的宫颈癌组织与癌旁正常组织,采用RT-qPCR法检测STING、CCL22、IDO表达。比较宫颈癌组织与癌旁正常组织STING、CCL22、IDO表达差异,分析宫颈癌组织STING、CCL22、IDO表达的关系及其与患者临床病理特征的关系,以及宫颈癌组织STING、CCL22、IDO表达与患者预后的关系。结果宫颈癌组织STING、CCL22、IDO相对表达量均高于癌旁正常组织(P均<0.05)。宫颈癌组织STING相对表达量与CCL22、IDO相对表达量均呈正相关关系(r分别为0.707、0.614,P均<0.05),CCL22相对表达量与IDO相对表达量亦呈正相关关系(r=0.601,P<0.05)。宫颈癌组织STING、CCL22、IDO相对表达量均与FIGO分期、组织分化程度有关(P均<0.05),而与年龄、病理类型、肌层浸润深度和淋巴结转移无关(P均>0.05)。以宫颈癌组织STING、CCL22、IDO相对表达量的均数为临界值,分别将患者分为STING、CCL22、IDO高表达者与低表达者。其中,STING高表达者41例、低表达者42例,CCL22高表达者40例、低表达者43例,IDO高表达者42例、低表达者41例。STING、CCL22、IDO高表达者3年总体生存率均低于其低表达者(χ^2分别为4.444、8.201、5.449,P均<0.05)。结论宫颈癌组织STING、CCL22、IDO高表达,其高表达与FIGO分期、组织分化程度和患者预后有关;三者表达变化可能协同促进宫颈癌的发生、发展。 Objective To observe the expression changes of stimulator of interferon gene(STING),CC chemokine ligand 22(CCL22),indoleamine-2,3-dioxygenase(IDO)in the cervical cancer and to explore its clinical significance.Methods The surgically removed cervical cancer tissues and adjacent normal tissues from 83 cases of patients with cervical cancer were selected.The expression levels of STING,CCL22 and IDO were detected by RT-qPCR.The differences in STING,CCL22 and IDO expression between the cervical cancer tissues and adjacent normal tissues were compared.The relationship between the expression of STING,CCL22,IDO and the clinicopathological features of cervical cancer,as well as the relationship between the expression of STING,CCL22,IDO and the prognosis of cervical cancer were analyzed.Results The relative expression levels of STING,CCL22 and IDO were higher in the cervical cancer tissues than in the adjacent normal tissues(all P<0.05).The relative expression of STING in the cervical cancer tissues was positively correlated with the relative expression of CCL22 and IDO(r=0.707,r=0.614,both P<0.05),while the relative expression of CCL22 was positively correlated with the relative expression of IDO(r=0.601,P<0.05).The relative expression levels of STING,CCL22 and IDO in the cervical cancer tissues were related to FIGO stage and tissue differentiation degree(all P<0.05),but not to age,pathological type,depth of myometrial invasion or lymph node metastasis(all P>0.05).According to the critical value which was the mean expression of STING,CCL22 and IDO in the cervical cancer tissues,the patients were divided into the high expression and low expression groups of STING,CCL22 and IDO,respectively.Among them,there were 41 patients with high expression of STING and 42 patients with low expression of STING,40 patients with high expression of CCL22 and 43 patients with low expression of CCL22,and 42 patients with high expression of IDO and 41 patients with low expression of IDO.The overall 3-year survival rate of patients with high expression of STING,CCL22 and IDO was lower than that of patients with low expression(χ^2=4.444,8.201 and 5.449,respectively,all P<0.05).Conclusion The high expression of STING,CCL22 and IDO in cervical cancer tissues is related to FIGO stage,tissue differentiation degree and patients'prognosis,which may synergetically promote the occurrence and development of cervical cancer.
