沉默BAK1、BCL2基因表达对HCC的HepG2细胞增殖、侵袭及转移的影响 被引量:1

摘要 目的:探讨沉默BAK1、BCL2基因表达对肝细胞肝癌(HCC)HepG2细胞增殖、侵袭及转移的影响。方法:将HCC HepG2细胞30株随机分为control组(不作转染处理)、NC-siRNA组(将NC-siRNA重组慢病毒质粒转染至HepG2细胞)、BAK1-siRNA组(将BAK1-siRNA重组慢病毒质粒转染至HepG2细胞)、BCL2-siRNA组(将BCL2-siRNA重组慢病毒质粒转染至HepG2细胞)、BAK1+BCL2组(将BAK1-siRNA、BCL2-siRNA重组慢病毒质粒转染至HepG2细胞),每组各6株;分别采用qRT-PCR法、CCK8法、Transwell小室侵袭实验、划痕实验检测转染后HepG2细胞BAK1和BCL2基因表达、增殖率、侵袭和转移能力。结果:BAK1+BCL2 siRNA组HepG2细胞中BAK1、BCL2 mRNA表达量、细胞增殖率、细胞迁移率、细胞侵袭能力低于BCL2-siRNA组、BAK1-siRNA组、NC-siRNA组、control组(P<0.05);BAK1+BCL2 siRNA组HepG2细胞增殖率随时间延长而明显降低(P<0.05)。结论:沉默BAK1、BCL2基因表达可抑制肝癌HepG2细胞细胞增殖及侵袭,促进其凋亡。 Objective: To investigate the effects of silencing BAK1 and BCL2 gene expression on proliferation,invasion and metastasis of hepatocellular carcinoma(HCC)HepG2 cells.Methods:Totally 30 HCC HepG2 cells were randomly divided into groups and received the corresponding treatments,namely,control group,NC-siRNA group,BAK1-siRNA group,BCL2-siRNA group and BAK1+BCL2 group,with 6 strains in each group.ThenqRT-PCR,CCK8,Transwell chamber invasion and scratch assay were used to detect the expression,proliferation,invasion and metastasis of BAK1 and BCL2 genes in HepG2 cells.Results:The mRNA expression,cell proliferation rate,cell migration rate and cell invasion ability of BAK1 and BCL2 in HepG2 cells were lowest in the BAK1+BCL2 siRNA group,followed by BCL2-siRNA group,BAK1-siRNA group,NC-siRNA group and control group(P<0.05).The proliferation rate of HepG2 cells in the BAK1+BCL2 siRNA group decreased significantly with time(P<0.05).Conclusion:Silencing the expression of BAK1 and BCL2 genes can inhibit the proliferation and invasion of HCC HepG2 cells and promote their apoptosis.
