摘要
通过设计不同富含G碱基的DNA序列,探究了G碱基对核酸-铜/银纳米簇(DNA-Cu/AgNCs)的荧光增强效应,并建立了铜离子的荧光检测方法。结果发现,在富含C碱基序列模板的5端增加G5序列后,制备得到的铜/银纳米簇的荧光强度显著增强。同时,该DNA-Cu/AgNCs的荧光可被Cu^2+和Hg^2+猝灭。通过NaBH 4掩蔽Hg^2+实现了对Cu^2+的特异性检测。该方法检测Cu^2+的线性范围为0.01~5.0μmol/L,检出限为5.0 nmol/L。方法具有简单快速、选择性高、成本低等优点,可用于实际样品测定。
A fluorescent method based on copper/silver nanoclusters for the detection of Cu^2+ was established by designing DNA sequences with G-rich bases and investigating the fluorescence enhancement effect of G base pairs on DNA-Cu/AgNCs.It was found that the fluorescence intensity of the prepared DNA-Cu/AgNCs was significantly enhanced by the G5 sequence at the 5'end of the C-rich sequence template.Meanwhile,the fluorescence of DNA-Cu/AgNCs was quenched by Hg^2+ and Cu^2+.NaBH 4 could be used to mask Hg^2+ in order to achieve a selective detection of Cu^2+.The linear range of this method for Cu^2+ was from 0.01μmol/L to 5.0μmol/L,and the detection limit was 5.0 nmol/L.The method is simple,rapid,highly selective and low-cost,and is able to apply in detection for real samples.
