茜草不同炮制品对子宫内膜出血模型大鼠的影响 被引量:7

Effects of Different Processed Medicine of Rubia Cordifolia L on Endometrial Hemorrhage Model Rats

摘要 目的:为了进一步探讨茜草、茜草炭的止血作用机制,通过建立大鼠子宫内膜出血模型,以研究茜草、茜草炭的复合提取物对凝血系统的影响及其止血机制。方法:空白组SD大鼠6只,灌胃给予5 g·L^-1羧甲基纤维素钠(sodium carboxymethyl cellulose,CMC-Na),造模SD大鼠20只灌胃给予戊酸雌二醇(2.5 mg·kg^-1),灌胃3周后,将造模成功的大鼠分为模型组、茜草复合提取物组及茜草炭复合提取物组,每组6只,空白组及模型组灌胃给予5 g·L^-1 CMC-Na,其他各给药组灌胃给予相对应的药物,7 d后观察茜草不同炮制品的复合提取物对子宫内膜出血模型大鼠子宫形态、以凝血四项试剂盒测定对凝血系统的影响、用酶联免疫吸附测定(enzyme linked immunosorbent assay,ELISA)法测定血栓素(thromboxane B2,TXB2)、6-酮-前列腺素F1α(6-keto-prostaglandin F1α,6-keto-PGF1α)的含量。结果:从形态上来看,模型组的子宫明显增厚且短小;与空白组比较,模型组明显缩短凝血酶时间(thrombin time,TT)、凝血酶原时间(prothrombin time,PT),延长活化部分凝血活酶时间(activated partial thromboplastin time,APTT),增加血浆纤维蛋白原(fibrinogen,FIB)水平(P<0.01)。与模型组比较,茜草与茜草炭复合提取物均能显著延长TT、PT,缩短APTT,降低FIB含量(P<0.01);茜草与茜草炭在缩短TT方面比较,差异有统计学意义(P<0.05);与空白组比较,模型组显著升高6-keto-PGF1α含量和TXB2含量(P<0.01);与模型组比较,茜草及茜草炭复合提取物均能够降低6-keto-PGF1α含量和TXB2含量(P<0.01),茜草与茜草炭比较,能够显著降低6-keto-PGF1α含量和TXB2含量(P<0.01),推测茜草相对于茜草炭对花生四烯酸代谢通路的影响更显著。结论:茜草、茜草炭通过作用于内源性、外源性凝血系统和花生四烯酸通路发挥显著的止血作用,而茜草炭对凝血系统的影响优于茜草,茜草较茜草炭对花生四烯酸通路的影响更显著。 Objective:To further explore the hemostatic mechanism of Qiancao(Rubia cordifolia L),carbonized Qiancao(Rubia cordifolia L).To study the effect of the compound extract of Qiancao(Rubia cordifolia L),carbonized Qiancao(Rubia cordifolia L)on the coagulation system and its hemostasis mechanism by establishing a rat model of endometrial hemorrhage.Methods:6 SD rats in the blank group were given 5 g·L^-1 sodium carboxymethyl cellulose(CMC-Na)by gavage,and 20 SD rats in the model group were given estradiol valerate(2.5 mg·kg-1)by gavage.After three weeks of gavage,the rats in the blank group and the model group were divided into model group,Qiancao(Rubia cordifolia L)compound extract group and carbonized Qiancao(Rubia cordifolia L)compound extract group.6 rats in each group were given 5 g·L^-1 CMC-Na by gavage.The corresponding drugs were given to each group by gavage.Seven days later,we observed the effects of the compound extracts of different processed medicine of Qiancao(Rubia cordifolia L)on the uterine morphology of the model rats with endometrial hemorrhage,measured the effects of four coagulation kits on the coagulation system,and measured the contents of thromboxane B2(TXB2)and 6-Keto-Prostaglandin F1α(6-keto-PGF1α)by enzyme linked immunosorbent assay(ELISA).Results:In terms of morphology,the uterus in the model group was significantly thickened and short;compared with the blank group,the model group significantly shortened the time of thrombin(TT),prothrombin(PT),prolonged the time of activated partial thromboplastin(APTT),and increased the content of fibrinogen(FIB)(P<0.01).Compared with the model group,the compound extract of Qiancao(Rubia cordifolia L)and carbon-ized Qiancao(Rubia cordifolia L)could significantly prolong TT and PT,shorten APTT and decrease FIB content(P<0.01),and there was significant difference in shortening TT between Qiancao(Rubia cordifolia L)and carbonized Qiancao(Rubia cordifolia L)(P<0.05);compared with the blank group,the content of 6-keto-PGF1αand TXB2 in the model group increased significantly(P<0.01);compared with the model group,the content of 6-keto-PGF1αand TXB2 in the composite extract of Qiancao(Rubia cordifolia L)and carbonized Qiancao(Rubia cordifolia L)decreased significantly(P<0.01);compared with carbonized Qian-cao(Rubia cordifolia L),Qiancao(Rubia cordifolia L)can significantly reduce the content of 6-keto-PGF1αand TXB2(P<0.01),suggesting that Qiancao(Rubia cordifolia L)has more significant effect on arachidonic acid metabolic pathway than car-bonized Qiancao(Rubia cordifolia L).Conclusion:Qiancao(Rubia cordifolia L)and carbonized Qiancao(Rubia cordifolia L)play a significant role in hemostasis by acting on the internal and external coagulation system and arachidonic acid pathway,while the effect of carbonized Qiancao(Rubia cordifolia L)on the coagulation system is better than that of Qiancao(Rubia cordifolia L),and the effect ofQiancao(Rubia cordifolia L)on arachidonic acid pathway is more significant than that of carbonized Qiancao(Rubia cordifolia L).
