摘要
目的:观察颗粒剂四君子膏和传统四君子膏对脾气虚模型大鼠胃肠功能、Ach E及NOS水平的影响。方法:60只SD大鼠(雌雄各半)随机分为正常组对照、模型对照组、颗粒剂四君子膏组(颗粒膏组)、传统四君子膏方组(传统膏组)、四君子汤组(汤剂组)。采用耗气破气+劳倦过度+饥饱失常法连续造模3周,复制脾气虚证大鼠模型,颗粒膏组、传统膏组、四君子汤组分别给予相应药物10 mL·kg^-1灌胃4周。第4周末,各组大鼠腹主动脉取血,取空肠组织,HE染色观察大鼠空肠组织病理形态学变化;通过酶联免疫法、免疫组织化学法检测各组大鼠乙酰胆碱酯酶(acetylcholin esterase,AchE)、一氧化氮合酶(nitric oxide synthase,NOS)水平;并计算胃残留率、小肠推进率。结果:与正常组比较,模型组大鼠血清及小肠组织AchE水平显著降低、模型组NOS水平显著升高、模型组大鼠胃残留率显著升高、模型组小肠推进率显著下降(P<0.05,P<0.01);经不同剂型四君子方治疗后,治疗组AchE水平显著上升、NOS水平显著下降,与模型组比较,差异均有统计学意义(P<0.05);颗粒膏组血清AchE、空肠NOS水平及小肠推进率与传统膏组比较,差异具有统计学意义(P<0.05)。结论:颗粒剂四君子膏方和传统四君子膏方两种剂型均可通过提高AchE水平,降低NOS活力改善大鼠胃肠平滑肌收缩和舒张运动,达到益气健脾的功效。因两种四君子膏剂型有差异,颗粒剂四君子膏可以通过提高AchE水平和小肠推进率,降低NOS活力起到益气健脾的功效。
Objective:To observe the effects of granules Sijunzi Cream and traditional Sijunzi paste on gastrointestinal function,AchE and NOS levels in spleen qi deficiency rats.Methods:Sixty SD rats(male and female)were randomly divided into normal group control,model control group,granule Sijunzi cream group(granular cream group),traditional Sijunzi Cream prescription group(traditional cream group),and Sijunzi decoction group(decoction).The method of Qi-brokend+over-exhaustion+hungersatisfaction was used to establish the model for 3 weeks,and the rat model of Spleen Qi deficiency syndrome was established.At the end of the fourth week,blood was taken from the abdominal aorta of each group of rats,jejunum tissue was taken,and the pathological and morphological changes of the jejunum tissue of the rats were observed by HE staining;acetylcholinesterase(AchE),nitric oxide synthase(NOS)levels;and calculated gastric residual rate and small intestinal advancing rate.Results:Compared with the normal group,the levels of AchE in serum and small intestine of the model group were significantly reduced,the NOS levels of the model group were significantly increased,the gastric residual rate of the model group was significantly increased,and the small intestine propulsion rate of the model group was significantly reduced(P<0.05,P<0.01);After treatment with Sijunzi prescription in different dosage forms,the AchE level in the treatment group increased significantly and the NOS level decreased significantly.Compared with the model group,the differences were statistically significant(P<0.05);the serum Ach E and jejunum in the granule group Compared with the traditional ointment group,the levels of NOS and small bowel advancing rate were statistically significant(P<0.05).Conclusion:Both granules Sijunzi cream and traditional Sijunzi Cream can improve the contraction and relaxation of gastrointestinal smooth muscle in rats by increasing the AchE level and reducing the activity of NOS.Due to the differences between the two Sijunzi Cream formulations,the granule Sijunzi Cream can improve the AchE level and the small intestine advancing rate,reduce the activity of NOS,and improve the qi and spleen.
