CBA/J小鼠虚热型反复自然流产模型的建立

Establishment of Recurrent Spontaneous Abortion with Deficiency Heat Syndrome in CBA/J Model Mice

目的:建立CBA/J小鼠虚热型反复自然流产模型。方法:将雌性CBA/J与雄性DBA/2J小鼠以2∶1合笼交配,制备反复自然流产模型。然后灌服附子、干姜、肉桂复方11 d,建立虚热型反复自然流产病症结合模型。观察每只小鼠的精神状态、皮毛颜色、舌象、大便性状并测定小鼠的肛温、食量、饮水量,与虚热证有关的生物化学指标、妊娠小鼠阴道出血情况、宫内胚胎发育、流产情况。结果:(1)虚热型反复自然流产组小鼠均于造模后出现精神萎靡、反应迟钝、毛色干枯、易怒、大便干结、饮水量明显增加等情况。(2)虚热型反复自然流产组小鼠T3、T4水平均高于正常组、反复自然流产组,差异具有统计学意义(P<0.05)。(3)虚热型反复自然流产组肛温较正常组、反复自然流产组明显增加,差异具有统计学意义(P<0.05)。均符合虚热证模型指标。(4)与正常组比较,反复自然流产组,虚热型反复自然流产组流产率明显增加,差异具有统计学意义(P<0.05)。结论:该模型可成为虚热型反复自然流产病症防治研究药效的实用模型。

Objective:To establish recurrent spontaneous abortion with deficiency Heat Syndrome in CBA/J model mice.Methods:Female CBA/J and male DBA/2J mice were mated in a 2∶1 cage to prepare a repeated spontaneous abortion model.Then,the compound of aconite,dried ginger and cinnamon was administered for 11 days,and a combined model of recurrent spontaneous abortion was established.Observe the mental state,fur color,tongue image,stool characteristics of each mouse,and measure the anus temperature,food intake and water consumption of the mice,biochemical indicators related to deficiency and heat syndrome,vaginal bleeding in pregnant mice,intrauterine embryos Development and abortion.Results:(1)The mice of the virtual-heat-type repeated spontaneous abortion group suffered from mental weakness,sluggish response,dry hair,irritability,dry stool,and markedly increased drinking water after modeling.(2)The levels of T3 and T4 in the hypothermia-type repeated spontaneous abortion group were higher than those in the normal group and repeated spontaneous abortion group,and the difference was statistically significant(P<0.05).(3)The anus temperature in the group of asthenia type of repeated spontaneous abortion increased significantly compared with the normal group and repeated spontaneous abortion group,the difference was statistically significant(P<0.05).All are in line with the index of deficiency heat syndrome model.(4)Compared with the normal group,the abortion rate in the repeated spontaneous abortion group of the virtual fever group increased significantly,and the difference was statistically significant(P<0.05).Conclusion:This model can be a practical model for the study of the efficacy of the research on the prevention and treatment of recurrent spontaneous abortion of asthenia type.
