三七粉对深静脉血栓模型大鼠血栓形成的预防作用 被引量:5

The Preventive Effect of Panax Notoginseng Powder on Thrombosis in Rats with Deep Venous Thrombosis

摘要 目的:分析三七粉对深静脉血栓模型大鼠的术后凝血功能紊乱及深静脉血栓形成的预防作用,为临床应用三七粉防治深静脉血栓提供实验依据。方法:96只SD大鼠按照随机数字表分为正常对照组、假手术组、模型组、三七粉组,除正常对照组6只外,其余每组各30只。对照组麻醉后处死,假手术组只游离下腔静脉,模型组、三七粉组用下腔静脉结扎法建立深静脉血栓模型。除正常对照组外,其余各组又按术后3 h、6 h、12 h、24 h、48 h共5个时间点分为5个亚组,每亚组6只,分别对5个时间点各个亚组大鼠抽取腹主动脉血液,ELISA测定大鼠血液的D-二聚体、纤维蛋白原、白细胞介素-6(interleukin-6,IL-6)、IL-8水平。结果:假手术组的D-二聚体、纤维蛋白原、IL-6、IL-8水平在手术后有一个快而短的升高趋势,在术后3 h时达到高峰,而后迅速回落。与假手术组比较,模型组各项指标在造模后3~24 h逐渐升高且各时间段亚组指标均高于假手术组,差异有统计学意义(P<0.05)。三七粉组与假手术组、模型组比较,同时段各个亚组的指标高于假手术组,差异有统计学意义(P<0.05或P<0.01)。结论:三七粉能够有效缓解大鼠血液的高凝状态,并能减轻相关的全身炎性反应,进而延缓大鼠深静脉血栓的发生发展。 Objective:To analyze the preventive effect of Sanqi Powder on the postoperative coagulation disorder and the formation of deep venous thrombosis in rats with deep venous thrombosis,so as to provide experimental basis for the clinical application of Sanqi Powder in the prevention and treatment of deep venous thrombosis.Methods:96 SD rats were divided into normal control group,sham operation group,model group and Sanqi Powder group according to the random number table,30 rats in each group except 6 in normal control group.The control group was killed after anesthesia,the sham operation group was only free of inferior vena cava,and the model group and Sanqi Powder group were used to establish the deep venous thrombosis model by ligation of inferior vena cava.In addition to the normal control group,the other groups were divided into 5 subgroups according to 3,6,12,24 and 48 hours after operation,6 rats in each subgroups.The blood of abdominal aorta was extracted from each subgroups at 5 time points.The contents of D-dimer,fibrinogen,interleukin-6(IL-6)and IL-8 were measured by ELISA.Results:The levels of D-dimer,fibrinogen,IL-6 and IL-8 in the sham operation group showed a fast and short increasing trend after operation,peaked at 3 hours after operation,and then fell back rapidly.Compared with the sham operation group,each index of the model group increased gradually in 3-24 hours after the model was made,and the index of the sub group in each time period was higher than that of the sham operation group,and the difference was statistically significant(P<0.05).Compared with the sham operation group and the model group,the indexes of each sub group of Sanqi Powder group were higher than those of sham operation group,and the difference was statistically significant(P<0.05 or P<0.01).Conclusion:Sanqi Powder can effectively alleviate the hypercoagulable state of blood and the related systemic inflammatory response in rats,and then delay the occurrence and development of deep venous thrombosis in rats.
