摘要
目的:利用网络药理学方法探究射干麻黄汤治疗支气管哮喘(简称哮喘)的分子作用机制。方法:利用中药系统药理数据库和分析平台(Traditional Chinese Medicines Systems Pharmacology Platform,TCMSP)数据库筛选出射干麻黄汤的有效化合物及化合物作用靶点,从TTD数据库和CTD数据库中检索哮喘相关的靶点,将二者进行交集得到射干麻黄汤治疗哮喘的潜在作用靶点,利用在线服务器及软件分析出射干麻黄汤治疗哮喘的关键作用靶点,然后利用生物信息学手段分析关键作用靶点参与的生物学过程和信号通路信息。结果:经过筛选后得到射干麻黄汤有效化合物114个,这些化合物共作用于79个靶点,其中与哮喘相关的靶点有39个,再次筛选后得到12个射干麻黄汤治疗支气管哮喘的关键作用靶点。这些作用靶点参与机体平滑肌细胞增殖的阳性调控、炎症反应、蛋白激酶B信号阳性调节等100种生物学过程及调控磷脂酰肌醇3-激酶(Phosphatidylinositol-3-Kinases,PI3K)/蛋白质丝氨酸苏氨酸激酶(Protein-serine-threonine Kinase,AKT)信号通路、MAPK信号通路、缺氧诱导因子-1(hypoxia-inducible factor,HIF-1)等45条信号通路。结论:本研究通过网络药理学方法初步揭示了射干麻黄汤对支气管哮喘的作用机制,射干麻黄汤通过黄芩素、黄芩苷、酸枣仁皂苷、儿茶素、槲皮素等多种有效成分,参与机体免疫反应、炎症反应等生物学过程和调控多种信号通路,以“多成分、多靶点、多途径”的特点干预哮喘疾病的发生发展,为下一步对射干麻黄汤治疗支气管哮喘的基础研究提供参考依据。
Objective:To explore the molecular mechanism of the the Shegan Mahuang decoction(射干麻黄汤)on bronchial asthma by means of network pharmacology.Methods:Effective compounds and targets of the Shegan Mahuang decoction were screened out by TCMSP database.The targets related to asthma were retrieved from TTD database and CTD database.Potential targets of the Shegan Mahuang decoction on asthma were obtained by intersecting the two databases.The key targets of the Shegan Mahuang decoction on asthma were analyzed by online servers and software.Bioinformatics was then used to analyze the key targets of the Shegan Mahuang decoction on asthma.Information technology is used to analyze the biological processes and signal pathway information involved by key targets.Results:After screening,114 effective compounds of the Shegan Mahuang decoction were obtained,which acted on 79 targets,of which 39 were related to asthma.After screening again,12 key targets of the Shegan Mahuang decoction on bronchial asthma were obtained.These targets are involved in 100 biological processes such as positive regulation of smooth muscle cell proliferation,inflammatory response,positive regulation of protein kinase B signal,and 45 signal pathways such as PI3K-Akt,MAPK and HIF-1.Conclusion:The mechanism of the Shegan Mahuang decoction on bronchial asthma is preliminarily revealed by network pharmacological method.the Shegan Mahuang decoction participates in biological processes such as immune response,inflammatory response and regulates multiple signaling pathways through baicalin,baicalin,jujube seed saponin,catechin,quercetin and other effective components.Characteristics of intervention in the occurrence and development of asthma disease,for the next step to the Shegan Mahuang decoction on bronchial asthma basic research to provide a reference basis.
