摘要
为了获得大通牦牛DQA2基因序列,并研究其序列特征及编码蛋白的结构和功能,本研究采用DNA混合池扩增后直接测序的方法,通过DNAstar软件和在线服务器预测大通牦牛DQA2基因CDS区编码蛋白的理化性质、疏水性/亲水性、糖基化位点、信号肽、跨膜结构、二级结构以及三级结构。结果表明,大通牦牛DQA2基因CDS区长度为765bp,编码253个氨基酸。蛋白结构预测结果表明,该蛋白是一种混合型的可溶性蛋白质,具有4个潜在的N-糖基化位点,分别为Asn31、Asn96、Asn260、Asn369;在第1~23位氨基酸之间存在信号肽;存在1个典型的跨膜螺旋区,氨基酸序号为216-239;蛋白质功能显示该蛋白存在信号受体活性、催化活性、运载体、调节代谢过程、细胞定位及免疫系统过程的功能。本研究结果为进一步研究大通牦牛的遗传育种提供理论基础。
In order to obtain the Datong Yak DQA2 gene sequence,and to study its sequence characteristics and the structure and function of the encoded protein.In this study,DNA sequencing software and online server were used to predict the physicochemical properties,hydrophobicity/hydrophilicity,glycosylation site,signal peptide and cross-section of the protein encoded by the CDS region of Datong Yak DQA2 gene.Membrane structure,secondary structure and tertiary structure.The results showed that the CDS region of Datong Yak DQA2 gene was 765 bp in length and encoded 253 amino acids.The protein structure predicted that the protein was a mixed soluble protein with four potential N-glycosylation sites.Asn31,Asn96,Asn260,Asn369;there is a signal peptide between amino acids 1 to 23;there is a typical transmembrane helix region with amino acid number 216-239;protein function shows signal receptor activity and catalysis The function of activity,carrier,regulation of metabolic processes,cell localization and immune system processes.The results of this study provide a theoretical basis for further research on the genetic breeding of Datong Yak.
