摘要
目的:探讨柴胡疏肝散对胃肠功能紊乱大鼠模型胃肠道Cajal间质细胞凋亡的影响。方法:将8周龄SD雄性大鼠随机分为sham组(对照组)、FGID组(模型组)、sham+药物组及FGID+药物组各15只。利用腹腔注射利血平建立胃肠功能紊乱大鼠模型,造模同时sham+药物组及FGID+药物组予柴胡疏肝散汤剂治疗,利用免疫细胞化学和TUNEL法检测柴胡疏肝散对模型大鼠胃肠道Cajal间质细胞凋亡的影响。结果:sham组内见较多Cajal间质细胞,极少见Cajal间质细胞凋亡;sham+药物组与sham组比较差异无统计学意义(P>0.05);FGID组Cajal间质细胞发生明显凋亡且细胞数量明显减少(P<0.05);FGID+药物组内Cajal间质细胞数量及凋亡情况较FGID组明显改善(P<0.05)。结论:柴胡疏肝散对正常大鼠Cajal间质细胞无影响,但能明显减轻胃肠功能紊乱模型大鼠胃肠道Cajal间质细胞的凋亡。
Objective:To discuss the effects of Chaihu Shugan powder on gastrointestinal interstitial cells of Cajal(ICCs)in rats with gastrointestinal dysfunction.Methods:SD male rats aged eight weeks were randomized into sham group(the control group),FGID group(the model group),sham+medicine group and FGID+medicine group,15 rats each group.Peritoneal injection of reserpine was used to establish rats model with gastrointestinal dysfunction,simultaneously,sham+medicine group and FGID+medicine group were treated by the decoction of Chaihu Shugan powder,immunocytochemistry and TUNEL method were adopted to detect the effects of Chaihu Shugan powder on the apoptosis of gastrointestinal ICCs.Results:More ICCs could be seen in sham group,scanty ICCs apoptosis could be seen;no significant difference could be found between sham+medicine group and sham group(P>0.05);in FGID group,ICCs occured the apoptosis notaly and the number of the cells reduced remarkably(P<0.05);the number and apoptosis conditions of ICCs in FGID+medicine group improved compared with FGID group remarkably(P<0.05).Conclusion:Chaihu Shugan powder shows no effects on ICCs of normal rats,but could obviously reduce the apoptosis of ICCs in rats with gastrointestinal dysfunction.
