香椿子多糖脱蛋白脱色工艺及其体外抗凝血活性研究 被引量:3

Study on Deproteinization and Decoloration Technology of Polysaccharide from Seeds of Toona Sinensis and Its Anticoagulant Activity in Vitro

摘要 研究香椿子多糖制备过程中脱蛋白脱色工艺及其体外抗凝血活性.以多糖保留率、蛋白质去除率和脱色率为指标,比较Sevag法、三氯乙酸(TCA)法、聚酰胺法对香椿子粗多糖的脱蛋白效果,采用活性炭法对脱蛋白后的多糖进行正交优化脱色,并对脱蛋白脱色后的多糖的抗凝血活性进行研究。研究表明,聚酰胺法脱蛋白效果最优,多糖保留率为88.09%,蛋白质去除率为68.15%;活性炭法脱色最优工艺为:活性炭用量2.0%,脱色温度60℃,脱色时间1 h,样液pH为4,在此工艺条件下多糖保留率达到80.23%,脱色率为64.58%。体外抗凝血实验表明,脱蛋白、脱色后的香椿子多糖能显著延长凝血酶原时间(Prothrombin Time,PT)和凝血酶时间(Thrombin Time,TT),对活化部分凝血活酶时间(Activated Partial Thromboplastin Time,APTT)影响不显著,表明其是通过影响外源性途径或共同途径发挥抗凝血作用。研究结果可为香椿子粗多糖纯化及抗凝血活性的研究提供基础。 The deproteinization and decolorization process in the preparation of polysaccharides from Seeds of Toona Sinensis(STS)and its anticoagulant activity in vitro were studied.Taking the polysaccharide retention rate,protein removal rate and the rate of decoloration as evaluation indexes,the deproteinization effects of Sevag method,TCA method and polyamide method on crude polysaccharide from STS were compared.Then,the activated carbon method was used to orthogonal optimize the deproteinized polysaccharide,and the anticoagulant activity of deproteinization and decolorized polysaccharide was studied.The results showed that the polyamide method had the best deproteinization effect,the retention rate of polysaccharide and the protein removal rate were reached to 88.09%and 68.15%.The optimal decolorization process of activated carbon were added 2.0%activated carbon at sample pH 4,with temperature at 60°C for 1h,repectively.Under the conditions,the retention rate of polysaccharide was 80.23%,and decolorization rate was reached to 64.58%.In vitro anticoagulation experiments showed that the polysaccharide from STS after deproteinization and decolorization could prolong PT and TT significantly,but had not significant effect on APTT.It was suggested the anticoagulant activity of STSP was worked by affecting the extrinsic coagulation system or common system.The results could provide a basis for the study of purification and anticoagulant activity of crude polysaccharides from Seeds of Toona sinensis.
