刺槐嫩枝扦插及生根酶活性的变化 被引量:9

Changes of Rooting Enzyme Activity in Cuttings and Shoots of Robinia pseudoacacia

摘要 以刺槐当年生嫩枝插条为材料,利用正交试验探讨扦插基质、无性系、植物生长调节物质的种类和浓度等因素对刺槐插穗生根的影响及不定根发生过程中吲哚乙酸氧化酶(IAAO)、过氧化物酶(POD)和多酚氧化酶(PPO)活性的变化及其与扦插生根之间的关系。结果表明,(1)试验因素对生根指标的影响排序为:无性系>激素浓度>激素种类,其中无性系山农选3号、用1500mg/L的IBA处理刺槐插穗基部的愈伤率达到86%,生根率达到75%,平均根长达到8.10cm,最大不定根长达到10.85cm,生根效果指数为4.42,为处理组中最优组合;(2)随着扦插时间的增加,PPO、POD和IAAO先增加再降低,在根源基诱导期最大,而激素种类和浓度的增加显著提高了酶活性增加和降低的幅度;(3)插穗酶活性对刺槐嫩枝扦插生根效果的影响排序为,IAAO>PPO>POD。 Taking the young branch cuttings of Robinia pseudoacacia as material,orthogonal test was used to study the effect of substratum,clones,kinds and concentrations of plant growth regulating substances on its cuttings rooting,and the relationship among the activity changes of indole acetic acid oxidase(IAAO),peroxidase(POD),polyphenol oxidase(PPO)during adventitious roots occurring process,and cuttings rooting.The results showed that:(1)The influence order of experimental factors on rooting index was:clone>hormone concentration>hormone type.With No.3 clone shannon treated with 1500mg/L IBA,the basal callus rate,rooting rate,average root length reached 86%,75%,and 8.10cm respectively,and the largest adventitious roots reached 10.85cm long,rooting effect index was 4.42,and it was the best combination.(2)With the increase of cutting time,the indicators of PPO,POD and IAAO showed a tendency of first increased at the beginning then decreased,and reached the maximum in the root group induction period,while the increase of hormone species and concentration significantly increased the range of enzyme activity increase and decrease.(3)The effect of enzyme activity of cuttings on the rooting of R.pseudoacacia twig cutting was ranked as:IAAO>PPO>POD.
