白喉棒状杆菌在实验教学中的染色方法改良

Staining improvement of Corynebacterium diphtheria in experimental teaching

目的探索实验教学中对白喉棒状杆菌的染色方法的改良。方法将复苏的白喉棒状杆菌接种于鸡蛋培养基上培养24 h,挑取菌体涂片,俾斯麦褐液染色1 min,后以奈瑟氏美蓝液染色1.5 min,制作得染色玻片。结果在油镜下可见清晰的蓝色菌体和明显的深蓝色异染颗粒。在较强及较弱的光线条件下菌体轮廓均可辨认。结论改良的染色方法可改进实验教学效果,其在临床上的适用性有待进一步研究。

Objective To reaserch the improvement of staining method of Corynebacterium diphtheriain experimental teaching.Method Cultured the resuscitated Corynebacterium diphtheriaon Egg-medium for 24 hours.Picked bacteria to smear slide,which was stained with Bismarck brown liquid for 1 minute,then with Methylene blue liquid for 1.5 min.Result Clear blue thallus and distinct dark blue metachromatic granule were seen in the oil objective len.Conclusion This improved staining method provided a better experimental teaching effect.The clinical applicability of this method remains to be farther studied.
