小柴胡汤对CFA大鼠血清细胞因子TNF-α、IL-1β、IL-6、M-CSF作用的实验研究

Experimental Study on Effect of Xiaochaihu Decoction on Serum Cytokines TNF-α,IL-1β,IL-6 and M-CSF in CFA Rats

目的基于治疗前后完全弗氏佐剂(CFA)大鼠血清细胞因子TNF-α、IL-1β、IL-6、M-CSF的表达,探讨小柴胡汤(XCHT)治疗类风湿关节炎的机制。方法动物实验将SPF级健康成年雌性Wistar大鼠应用弗氏完全佐剂和牛Ⅱ型胶原制备大鼠类风湿关节炎CFA动物模型。根据随机数字表,将大鼠随机分为正常组、模型组、XCHT低剂量组、XCHT中剂量组、XCHT高剂量组、雷公藤多苷组。造模后分别按照药物干预方案, 2 mL/次,日3次灌胃,实验观察大鼠一般状态、关节炎状态及酶联免疫吸附测定大鼠血清中细胞因子TNF-α、IL-1β、IL-6、M-CSF的含量。结果大鼠随着小柴胡汤剂量的增加,一般状态及关节炎状态改善。在血清炎性细胞因子TNF-α、IL-1β、IL-6、M-CSF检测方面,随着小柴胡汤剂量的增大,其表达逐渐降低,与模型组比较,小柴胡汤中及高剂量统计结果显示P<0.01,降低很显著。结论本研究显示小柴胡汤随着剂量的增加,可以改善CFA大鼠的一般状态及关节炎状态。小柴胡汤能够通过抑制炎性细胞因子TNF-α、IL-1β、IL-6、M-CSF的表达,抑制炎症反应,从而延缓骨破坏,为中药方剂治疗类风湿关节炎提供基础实验依据。

Objective This study was to explore the mechanism of Xiaochaihu Decoction in the treatment of rheumatoid arthritis based on the expressions of serum cytokines TNF-α,IL-1β,IL-6 and M-CSF in CFA rats before and after treatment. Methods In animal experiments, we applied SPF health adult female Wistar rats for the preparation of CFA animal model of rats with rheumatoid arthritis with freund’s complete adjuvant and cattle Ⅱ collagen type.According to the random number table,the rats were randomly divided into the normal group,the model group,the Xiaochaihu Decoction low-dose group,the Xiaochaihu Decoction medium-dose group,the Xiaochaihu Decoction high-dose groupand the Tripterygium Glucosides group.After the model was built, the rats were given to fill the stomach respectively three times a day, 2 mL/time, according to the drug intervention program.We observed the general state,the state of arthritis in rats and determined the contents of cytokines TNF-α,IL-1β,IL-6 and M-CSF in rats serum by enzyme-linked immunosorbent assay.Result With the increase of the dosage of Xiaochaihu Decoction, the general state and arthritis state of rats were improved.In the examination of serum levels of inflammatory cytokine TNF-α,IL-1β,IL-6 and M-CSF, with the increase of Xiaochaihu Decoction dose, their expressions gradually decreased. Compared with the model group,the Xiaochaihu Decoction medium-dose group and the Xiaochaihu Decoction high-dose group statistical results showed significant difference(P<0.01).Conclusion This study shows that as the dose of Xiaochaihu Decoction increases,it could improve the general state of CFA rats and the state of arthritis.Xiaochaihu Decoction might inhibit inflammation by inhibiting inflammatory cytokines TNF-α,IL-1β,IL-6 and M-CSF to delay bone destruction.It would provide the basic experimental basis for the treatment of rheumatoid arthritis of traditional Chinese medicine.
