宫颈癌组织中BIM和Girdin蛋白表达与临床病理特征及预后的相关性分析 被引量:5

摘要 目的分析宫颈癌组织中Bcl-2相互作用细胞凋亡调节因子(Bcl-2 interacting mediator of cell death,BIM)和Girdin蛋白的表达情况,并探讨BIM和Girdin蛋白与宫颈癌患者临床病理特征及预后的相关性。方法选取延安大学附属医院2011年6月-2013年6月收治的136例宫颈癌患者作为研究对象,采用系统回顾性分析法分析所有患者的临床及随访资料,收集宫颈癌患者其136份癌组织标本(研究组)及136份其他良性宫颈疾病的正常组织标本(对照组)作为研究对象,所有组织标本均行BIM与Girdin蛋白的检测,分析宫颈癌组织中BIM与Girdin蛋白的表达与患者临床病理特征和预后的关系。结果宫颈癌组织中BIM及Girdin蛋白阳性表达率分别为36.03%、58.09%(P<0.05);BIM及Girdin蛋白的表达与宫颈癌患者年龄、病理类型差异均无统计学意义(P>0.05);BIM及Girdin蛋白的表达与宫颈癌患者淋巴结转移、FIGO分期、分化程度及肌层浸润深度差异有统计学意义(P<0.05);经单因素分析得出:FIGO分期、分化程度、肌层浸润深度、BIM蛋白及Girdin蛋白患者其5年生存率率差异有统计学意义(P<0.05);多因素COX比例风险回归模型分析显示,FIGOⅡ期、低分化、肌层浸润深度≥1/2、BIM蛋白阴性及Girdin蛋白阳性均为影响宫颈癌患者预后的独立危险因素(P<0.05)。结论在宫颈癌癌变组织中BIM呈低表达,Girdin蛋白呈高表达,随着患者FIGOⅡ期、低分化、肌层浸润深度≥1/2时BIM低表达率和Girdin蛋白高表达率均上升,且联合FIGOⅡ期、低分化、肌层浸润深度≥1/2共同为影响宫颈癌患者预后的独立危险因素。 Objective To analyze the expressions of BIM and Girdin protein in cervical cancer tissues,and to explore the correlations between BIM and Girdin protein on one hand and clinicopathological features and prognosis of patients with cervical cancer on the other hand.Methods One hundred and thirty-six patients with cervical cancer admitted to the Affiliated Hospital of Yan’an University between June 2011 and June 2013 were selected.The clinical and follow-up data of all these patients was analyzed retrospectively.136 cancer tissue specimens(study group)and 136 other normal tissue specimens of benign cervical diseases(control group)were selected as the subjects.BIM and Girdin proteins were detected in all the tissue samples,and the relationships between the expressions of BIM and Girdin protein in cervical cancer tissues and clinicopathological features and prognosis were analyzed.Results The positive expression rates of BIM and Girdin protein in cervical cancer tissues were 36.03%and 58.09%,respectively(P<0.05).The difference in expressions of BIM and Girdin protein between patients of different age and pathological types of cervical cancer was not statistically significant(P>0.05).However,the difference in expressions of BIM and Girdin protein between patients with or without lymph node metastasis,with different FIGO stage,differentiation degrees and depths of myometrial invasion was statistically significant(P<0.05).Univariate analysis showed that the difference in the FIGO stage,degree of differentiation,depth of muscular invasion,expressions of BIM protein and Girdin protein and five-year survival rates was statistically significant(P<0.05).The multivariate Cox proportional hazards regression model showed that FIGOⅡstage,poor differentiation,depth of myometrial invasion≥1/2,BIM that tested negative and Girdin protein that tested positive were independent risk factors for the prognosis of patients with cervical cancer(P<0.05).Conclusion The expression of BIM is low but Girdin protein is highly expressed in cervical cancerous tissues.With the FIGO stageⅡ,poor differentiation,and depth of myometrial invasion≥1/2,the low expression rate of BIM and the high expression rate of Girdin protein can increase,so the combination of FIGOⅡstage,poor differentiation,and depth of muscle infiltration≥1/2 is an independent risk factor for prognosis in patients with cervical cancer.
