摘要
目的:探讨了骨髓间充质干细胞(BMSC)移植对丙烯酰胺中毒大鼠周围神经病的治疗效果和可能的作用机制。方法:60只SD大鼠随机分为对照组、模型组和BMSC组,模型组采用丙烯酰胺灌胃大鼠构建中毒性周围神经病模型,建模成功后,BMSC组大鼠经尾静脉注射BMSC 1×107个,对照组和模型组注射等体积的生理盐水。分别在治疗后第0、1、2、3、4和5周分析3组大鼠步态变化;第5周坐骨神经传导潜伏期和神经传导速度的变化;染色分析坐骨神经病理程度,Western blot分析每组大鼠周围神经组织神经生长因子(NGF)和胶质细胞源性神经生长因子(GDNF)水平;分析每组大鼠氧化应激水平。结果:与模型组比,BMSC组大鼠治疗后各时间点步态评分明显降低(P<0.05),神经传导潜伏期明显降低(P<0.05),神经传导速度显著增加(P<0.05)。与模型组比,BMSC组大鼠坐骨神经病变病理评分显著下降(P<0.05),坐骨神经组织NGF和GDNF等蛋白水平显著增加(P<0.05),外周血中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)和过氧化氢酶(CAT)水平显著增加(P<0.05)。结论:BMSC可显著提高丙烯酰胺中毒大鼠的神经组织营养因子和抗氧化应激水平,进而改善了中毒大鼠的周围神经病变程度。
Objective:To investigate the therapeutic effect and possible mechanism of bone mesenchymal stem cells(BMSC)transplantation on peripheral neuropathy in rats with acrylamide poisoning.Methods:60 SD rats were randomly divided into 3 groups:the control group,the model group and the BMSC group.The model group was established by intragastric administration of acrylamide to toxic peripheral neuropathy.The gait changes,sciatic nerve conduction latency and nerve conduction velocity were analyzed at 0,1,2,3,4 and 5 weeks after treatment.The pathological changes of sciatic nerve were analyzed by staining.The levels of nerve growth factor(NGF)and glial cell line-derived neurotrophic fcator(GDNF)in peripheral nerve tissue were analyzed by Western blot.The levels of oxidative stress were analyzed.Results:Compared with the model group,the gait score and nerve conduction latency of the BMSC group had significantly decreased at each time point after treatment(P<0.05),the nerve conduction velocity significantly increased(P<0.05),the pathological score of sciatic neuropathy significantly decreased(P<0.05),the levels of NGF and GDNF in the sciatic nerve tissue,the levels of SOD(superoxide dismutase),glutathione peroxidase(GSH-Px)and CAT(catalase)in peripheral blood of BMSC group significantly increased(P<0.05).Conclusion:BMSC can significantly increase the levels of neurotrophic fcators and antioxidant stress in rats with acrylamide poisoning,thereby relieving the peripheral neuropathy in rats with acrylamide poisoning.
