摘要
[目的]探讨独角石斛组织培养及快繁技术,为其规模化生产提供技术支持。[方法]以独角石斛侧芽作为外植体,应用正交试验等方法,对不定芽诱导、增殖、壮苗生根培养及组培苗移栽等进行优化。[结果]独角石斛侧芽可诱导出不定芽,以MS+2.0 mg·L-1 6-BA+0.1 mg·L-1 NAA+30 g·L-1蔗糖+7.5 g·L-1琼脂为培养基,不定芽诱导效果最好;丛生芽最适继代增殖培养基为:MS+2.0 mg·L-1 6-BA+0.5 mg·L-1 KT+0.4 mg·L-1 NAA+30 g·L-1蔗糖+7.5 g·L-1琼脂,增殖系数达4.78;丛生芽最适生根培养基为:1/2 MS+0.4 mg·L-1 NAA+0.5 mg·L-1 IBA+0.5 g·L-1 AC+50 g·L-1土豆+50 g·L-1香蕉泥+20 g·L-1蔗糖+7.0 g·L-1卡拉胶,生根率达100%,苗生长健壮;组培苗经15 d炼苗后移栽到水苔基质中,45 d后成活率达90.5%。[结论]通过侧芽直接诱导不定芽,可以建立独角石斛高效快繁技术体系。
[Purpose] To provide technical support for large-scale production of Dendrobium unicorneum, we sought a rapid propagation system of D.unicorneum. [Method] Developmental processes of adventitiou shoot induction, propagation, root induction, and transplanting were optimized using D.unicorneum lateral bud as the explant by methods such as othogonal design. [Result] The most effective adventitious buds differentiation was achieved on the MS medium supplemented with 2.0 mg·L-1 6-BA, 0.1 mg·L-1 NAA, 30 g·L-1 sucrose and 7.5 g·L-1 agar. The optimal MS medium for maximum shoot number(4.78) is to supplement 2.0 mg·L-1 6-BA, 0.5 mg·L-1 KT, 0.4 mg·L-1 NAA, 30 g·L-1 sucrose and 7.5 g·L-1 agar. A robust seedling with 100% rooting rate can be achieved on the medium supplemented with1/2 MS, 0.4 mg·L-1 NAA, 0.5 mg·L-1 IBA, 0.5 g·L-1 AC, 50 g·L-1 potato, 50 g·L-1 banana mash, 20 g·L-1 sucrose and 7.0 g·L-1 carrageenan. A survival rate of 90.5% can be achieved by growing previously hardened tissue-cultured seedling in water moss matrix for 45 d.[Conclusion] A rapid and effective propagation system of D.monolobium can be established by using adventitious bud of D.monolobium induced by lateral shoot.
