乳酸杆菌产细菌素对金黄色葡萄球菌和肺炎链球菌临床株的体外抑菌效果研究 被引量:2

Bacteriostatic effect of lactic acid bacillus on clinical strains of Staphylococcus aureus and Streptococcus pneumoniae

摘要 目的分析乳酸杆菌产细菌素对两种临床致病菌的抑菌效果。方法选择2016年2月-2017年4月肺部感染患者10例为研究对象,从痰液中各提取5株金黄色葡萄球菌及5株肺炎链球菌作为测试菌株。将乳酸杆菌产细菌素与两种致病菌混合培养,对菌落进行计数,并通过扫描电镜观察生物膜形成情况,评估乳酸杆菌产细菌素对致病菌的抑制效果。结果乳酸杆菌产细菌素对金黄色葡萄球菌及肺炎链球菌生长具有显著的抑制效果。细菌素对5株金黄色葡萄球菌表现出明显的抑菌效果(P<0.05),其抑菌效率均>70%。而细菌素对5株肺炎链球菌中的4株表现出较强的抑菌效果(P<0.05),其抑菌效率均>65%。细菌素对5株金黄色葡萄球菌中的4株生物膜的形成表现出较强的抑制效果(P<0.05),并且细菌素显著减少了5株肺炎链球菌的4株生物膜的细菌数量(P<0.05),抑菌效果较好的菌株中,细菌生长数量低于对照组。结论乳酸杆菌产细菌素对金黄色葡萄球菌及肺炎链球菌的生长及生物膜表现出较强的抑制效果,提示乳酸杆菌产细菌素对于治疗临床肺部感染具有重要意义。 OBJECTIVE This study was designed to investigate the bacteriostatic effects of lactic acid bacillus on two strains of clinical pathogens.METHODS Ten patients with pulmonary infection were studied between Feb.2016 to Apr.2017,and 5 strains of Staphylococcus aureus and Streptococcus pneumoniae were isolated as the test strains from sputum.The bacteriostatic effects of lactic acid bacillus on the growth and biofilm formation of two clinical pathogen strains were observed by colony counting and scanning electron microscopy in the co-cultured medium of bactirocin and two pathogens.RESULTS The bacteriocin produced by lactic acid bacillus had significant antibacterial effect on the growth of S.aureus and S.pneumoniae.Bacteriocin exhibited significant antibacterial effect on 5 strains of S.aureus(P<0.05),and the bacteriostatic efficiency was higher than 70%,whereas the bacteriocin significantly inhibited 4 in 5 strains of S.pneumoniae(P<0.05)and the bacteriostatic efficiency was higher than 65%.Bacteriocins showed strong inhibitory effect on the formation of biofilms of 4 in 5 strains of S.aureus(P<0.05),and bacteriocins significantly reduced the number of bacteria of 4 biofilms in 5 strains of S.pneumoniae(P<0.05).Among the strains with better bacteriostatic effect,the bacteria numbers was lower than that of control.CONCLUSION The bacteriocin produced by lactic acid bacteria exhibited significantly bacteriostatic effect on the growth and biofilm formation of S.aureus and S.pneumoniae in vitro,suggesting the significance of bacteriocin produced by lactic acid bacillus in the treatment of clinical pulmonary infection.
