电针干预对功能性消化不良大鼠下丘脑CRF及其受体的影响 被引量:8

Effect of Electroacupuncture on CRF and Its Receptor in Hypothalamus of Rats with Functional Dyspepsia

摘要 目的观察电针干预对功能性消化不良(FD)大鼠焦虑行为的影响,检测下丘脑CRF及其受体表达情况,以探究电针干预FD的作用机制。方法将36只大鼠随机分为正常组(12只)、造模组(24只)。采用复合因素(改良夹尾法+0℃生理盐水灌胃+不规则饮食法)干预建立FD模型。造模后随机选取2只大鼠,解剖后无器质性病变,表明造模成功。将造模成功的大鼠随机分为模型组(11只),电针组(11只)。电针组予以双侧"足三里"穴,"太冲"穴针刺后接电针。每日1次,连续干预14 d。干预结束后行旷场试验,并采用Western blotting检测下丘脑CRF、CRF-R1、CRF-R2蛋白表达量。结果与正常组比较,模型组大鼠水平运动格数、垂直运动次数及中央格停留时间均显著减少(P<0.01);下丘脑CRF、CRF-R1表达增多,CRF-R2表达减少(P<0.01)。与模型组相比较,电针组大鼠水平运动格数、垂直运动次数及中央格停留时间显著增加(P<0.01)。下丘脑CRF、CRF-R1表达减少,CRF-R2表达增多(P<0.01)。结论电针干预可减轻FD大鼠焦虑样行为,其作用机制可能是通过调节下丘脑CRF及其受体的表达实现的,且CRF-R1与CRF-R2之间的平衡与之具有相关性。 Objective:To observe the effect of electroacupuncture on anxiety behavior of functional dyspepsia rats,and the expression of hypothalamic CRF and its receptor,and to explore its mechanism.Methods:36 rats were randomly divided into the normal group(12 rats)and the modeling group(24 rats).FD model was established by intervention of multiple factors.The rats were randomly divided into the model group and electroacupuncture group,11 rats in each.In the electroacupuncture group,Zusanli points and Taichong points were electroacupunctured once a day for 14 days.After the intervention,the open field test was carried out,and the expression of CRF,CRF-R1 and CRF-R2 in hypothalamus was detected with Western blotting.Results:Compared with the normal group,the number of horizontal movement,vertical movement and central stay time in the model group were significantly reduced(P<0.01);the expression of CRF and CRF-R1 in hypothalamus increased,and the expression of CRF-R2 decreased(P<0.01).Compared with the model group,the number of horizontal movement,the number of vertical movement and the time of staying in the central grid in the electroacupuncture group were significantly increased(P<0.01).The expression of CRF and CRF-R1 in hypothalamus decreased,and the expression of CRF-R2 increased(P<0.01).Conclusion:Electroacupuncture can alleviate anxiet-like behaviors in rats with functional dyspepsia,which may be achieved by regulating the expression of CRF and its receptor in the hypothalamus,and the balance between CRF-R1 and CRF-R2 is correlated with it.
