五苓散对肾阳虚型口干燥症小鼠颌下腺AQP-5的分布表达研究 被引量:3

摘要 目的通过研究五苓散对肾阳虚模型小鼠颌下腺AQP-5分布表达的作用,探究五苓散治疗肾阳虚型小鼠口干燥症的机制。方法将27只ICR小鼠随机分为空白组、模型组、五苓散组,除空白组外,其余各组采用肌肉注射氢化可的松诱导联合限制日摄食量法建立气虚基础上的肾阳虚模型。造模后给予相应的药物干预10d,空白组、模型组均灌胃0.2mL/10g蒸馏水,五苓散组以每日0.2mL/10g灌胃五苓散汤剂,观察给药后的小鼠一般情况,测量小鼠的饮水量、唾液流量,检测颌下腺形态及AQP-5分布表达。结果与空白组相比,模型组出现颌下腺萎缩,饮水量减少,唾液流量减少,AQP-5主要表达在基底膜和顶膜中,较空白组染色较弱;相比于模型组,五苓散组颌下腺萎缩减少,饮水量增加,唾液流量,并且一定程度上AQP-5表达分布染色较模型组强。结论五苓散能抑制颌下腺萎缩,增加饮水量和唾液流量,可能通过改变AQP-5的分布表达改善小鼠在气虚基础上的肾阳虚证引起的口干症状。 Objective:To investigate the effect of Wuling Powder on the expression of AQP-5 in the submandibular gland of mice with kidney Yang deficiency model,and to explore the mechanism of Wuling Powderon xerostomia.Methods:27 ICR mice were randomly divided into the blank group,the model group and Wuling Powder group.Except the blank group,the other groups established Qi deficiency and Yang deficiency model by hydrocortisone induction combined with limited daily intake.After 10 days of drug intervention,the blank group and the model group were given 0.2 mL/10 g normal saline,and Wuling Powder group was given 0.2 m L/10 g Wuling Powder daily.The general situation of mice after administration was observed.The water consumption and saliva flow of mice were measured,and the morphology of submandibular gland and the expression of AQP-5 distribution were detected.Results:Compared with the blank group,the model group showed submandibular gland atrophy,reduced water consumption,reduced salivary flow,and AQP-5 was mainly expressed in the basement membrane and parietal membrane,which was weaker than the blank group.Compared with the model group,Wuling Powder group showed less submandibular gland atrophy,increased water consumption and increased salivary flow,which alleviated dry mouth,and for a certain period of time.The degree of staining in AQP-5 expression distribution was stronger than that of model group.Conclusion:Wuling Powder can inhibit submandibular gland atrophy,increase water consumption and salivary flow.It may improve xerostomia caused by kidney Yang deficiency in mice by altering the expression of AQP-5.
