宣肺通腑方加减对肠缺血/再灌注肺损伤导致的急性肺损伤大鼠抗炎、抗氧化作用

Anti-inflammatory and Anti-oxidative Effects of Xuanfei Tongfu Prescription on Rats with Acute Lung Injury Induced by Intestinal Ischemia/Reperfusion

目的研究宣肺通腑方加减对肠缺血/再灌注肺损伤导致的急性肺损伤大鼠抗炎、抗氧化作用。方法选取40只大鼠,假手术组大鼠10只,制作肠缺血/再灌注肺损伤模型,建模成功,随机分为模型组、地塞米松组、宣肺通腑方组各10只。地塞米松组给予地塞米松,宣肺通腑方组给予宣肺通腑方,假手术组和模型组给予生理盐水。检测用力肺活量(FVC)、肺总量(TLC)、第1秒时间肺活量(FEV1)及1秒率(FEV1/FVC)检测,白细胞介素-8(IL-8)、肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、血管内皮生长因子(VEGF)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、一氧化氮合酶(NOS)及髓过氧化物酶(MPO)水平及p38、p-p38、pNF-κB蛋白表达。结果与模型组比较,宣肺通腑方组FVC、TLC、FEV1、FEV1/FVC、IL-8、TNF-α、IL-6及VEGF水平降低(P<0.05);SOD、GSH-Px水平升高,NOS、MPO水平降低(P<0.05),p38、p-p38及p-NF-κB蛋白降低(P<0.05)。结论宣肺通腑方加减对肠缺血/再灌注肺损伤导致的急性肺损伤大鼠作用明显,能有效改善大鼠肺功能,通过调控p38/NF-κB通路,降低IL-8、TNF-α、IL-6及VEGF而发挥抗炎作用,提高SOD、GSHPx水平,降低NOS、MPO水平,起到治疗急性肺损伤的作用。

Objective:To study the anti-inflammatory and antioxidant effects of Xuanfei Tongfu Decoction on acute lung injury rats induced by intestinal ischemia/reperfusion lung injury.Methods:The model of intestinal ischemia/reperfusion lung injury in SD rats was established successfully.The rats were randomly divided into the model group,dexamethasone group,Xuanfei Tongfu Decoction group and sham operation group.Dexamethasone group was given intraperitoneal injection of dexamethasone;Xuanfei Tongfu Decoction group was given water decoction of Xuanfei Tongfu Decoction;sham operation group and the model group were given normal saline intragastric administration.FVC,TLC,FEV1 and FEV1/FVC,IL-8,TNF-α,IL-6,VEGF,SOD,the levels of GSH-Px,NOS,MPO and the expression of p38,p-p38 and p-NF-κB proteins were measured.Results:Compared with the model group,the levels of FVC,TLC,FEV1,FEV1/FVC,IL-8,TNF-α,IL-6 and VEGF decreased(P<0.05);the levels of SOD and GSH-Px increased;the levels of NOS and MPO decreased(P<0.05);the levels of p38,p-p38 and p-NF-κB decreased in Xuanfei Tongfu Decoction group(P<0.05).Conclusion:Xuanfei Tongfu Decoction has obvious effects on acute lung injury rats caused by intestinal ischemia/reperfusion lung injury.It can effectively improve the lung function of rats.By regulating p38/NF-κB pathway,it can reduce IL-8,TNF-α,IL-6 and VEGF,play an anti-inflammatory role,improve SOD and GSH-Px levels,reduce NOS and MPO levels,and play an antioxidant role.
