转iaaM基因黄花蒿中基因表达量与其腺毛密度相关性分析

摘要 以转化腺毛中特异表达iaa M基因的13株黄花蒿单株为材料,用半定量RT-PCR法对转基因各单株中转基因的表达进行定量,并利用荧光显微镜对这些转基因个体叶片腺毛的密度进行观察。结果:iaa M基因在转基因黄花蒿植株中均有表达,基因表达量与腺毛密度呈正相关,其中表达量最高的转基因植株个体iaa M-2腺毛密度达到59.9枚/mm^2,为对照的2倍以上,表明腺毛特异启动子调控的iaa M基因转化黄花蒿,能显著促进黄花蒿腺毛的发育,增加其腺毛密度。由于黄花蒿腺毛是其有效成分青蒿素的主要合成和贮存部位,可望通过这种特异的转基因手段有效提高黄花蒿中青蒿素的含量。 The 13 individual transgenic plants of Artemisia annua with specific expression of iaa M gene in its glandular trichome are used as materials in this study.The gene expression was quantified by semi-quantitative RT-PCR method and the density of glandular trichome in the leaves of these transgenic individuals was observed by fluorescence microscope.The results showed that iaa M gene was expressed in all the transgenics.The gene expression level was positively correlated with the density of its glandular trichome.Among the transgenic individuals iaa M-2 with the highest density 59.9/mm^2 which are more than 2 times to the control accords the highest expression level.The results showed that the transformation of iaa M gene under the regulation by the specific promoter of glandular trichome can significantly promote the development of glandular trichome and increase the glandular density in Artemisia annua.Since the glandular trichome is the main synthesis and storage cells of artemisinin,it is expected to effectively increase the content of artemisinin in Artemisia annua through this specific transgene.
