摘要
目的通过测定发酵冬虫夏草菌粉中虫草多糖(CSP)的含量及建立辐射损伤动物模型,初步探讨虫草多糖(CSP)的抗辐射作用。方法采用热水浸提法提取发酵冬虫夏草菌粉中的多糖并利用改良的苯酚硫酸法测定虫草多糖的含量。利用医用电子直线加速器对小鼠进行全身一次性X射线照射,观察存活率,检测外周血象,计算胸腺及脾脏脏器指数。结果发酵冬虫夏草菌粉中多糖的平均含量为1.35%,改良后的苯酚硫酸法所测吸光度、精密度和重复性良好,测定结果准确可靠。虫草多糖能提高电离辐射损伤小鼠的存活率和存活时间;加速白细胞的恢复,但对红细胞、血红蛋白、血小板无显著影响;提高胸腺及脾脏的脏器指数。虫草多糖对骨髓DNA含量的影响无统计学意义。结论发酵冬虫夏草中的多糖是冬虫夏草发挥辐射防护作用的物质基础,对X射线辐射损伤小鼠具有一定的保护作用。
Objective To investigate the protective effect of Cordyceps sinensis polysaccharides(CSP)against radiation in mice through the assay of polysaccharides in fermented Cordyceps sinensis and the establishment of the animal model of ionizing radiation damage.Methods The polysaccharides from fermented cordyceps sinensis powder was extracted by hot water and its content was determined by modified phenol-sulfuric acid method.Mice were exposed to total-body X-ray radiation using a medical linear accelerator.The survival rate of mice,blood cell counts,immune organ index(thymus and spleen),DNA contents of bone marrow cell were observed after radiation.Results The average content of polysaccharides in the fermented Cordyceps sinensis was 1.35%.The improved phenol-sulfu-ric acid method showed stable absorbance,good precision and repeativeness,and the results are accurate and relia-ble.Cordyceps sinensis polysaccharides improved the survival rate and time in radiation-injured mice,and acceler-ated the recovery of WBC,but showed no significant effects on RBC,HGB,and PLT,and increased the organ index of thymus and spleen.Conclusion Polysaccharides in fermented Cordyceps sinensis are the material basis for Cordyceps sinensis to play a role in protection against X-ray radiation.
