高效液相色谱法同时测定蟾酥中8种成分的含量

Simultaneous determination of 8 components in Bufonis Venenum by HPLC

目的建立HPLC法同时检测蟾酥中华蟾酥毒基、酯蟾毒配基、日蟾毒它灵、蟾毒灵、沙蟾毒精、蟾毒它灵、远华蟾毒精和华蟾毒它灵8种成分的含量。方法采用Thermo Scientific C18色谱柱(250mm×4.6 mm,5μm),流动相为乙腈-水,梯度洗脱,检测波长296 nm,流速0.6 mL·min^-1,柱温30℃,进样量10μL。结果8个成分在对应线性浓度范围内与峰面积线性关系良好,精密度、稳定性和重复性的RSD均<5%,平均回收率在97.2%~99.8%。结论该含量测定方法操作简便、准确可靠,可用于蟾酥的质量控制。

Objective To simultaneously determine cinobufagin,resibufogenin,gamabufalin,bufalin,arenobufagin,bufotalin,telocinobufagin,and cinobufotalin in Bufonis venenum by HPLC.Methods The separation was performed on Thermo Scientific C18(250 mm×4.6 mm,5μm)column.The mobile phase was consisted of acetonitrile and water and the flow rate was 0.6 mL·min^-1.The detection wavelength was 296 nm,the column temperature was 30℃,and the sample size was 10μL.Results All the compounds showed good linearity in the corresponding range of concentration.The RSD of the precision,reproducibility and stability was less than 5%.The average recovery ranged from 97.2%to 99.8%.Conclusion The method is accurate and simple,which can be used for the quality control of Bufonis venenum.
