超高效合相色谱法测定胃瘫外敷方中厚朴酚与和厚朴酚的含量的研究及其影响因素分析 被引量:2

摘要 目的建立超高效合相色谱(UPC2)测定胃瘫外敷方中厚朴酚、和厚朴酚的含量,探讨UPC2使用时的主要影响因素。方法采用Waters Acquity UPC2 BEH色谱柱(150 mm×2.1 mm,1.7μm);流动相为A为CO2,B为甲醇,梯度洗脱条件:0~3 min:2%~10%B;3~4 min:10%~35%B;4~8min:35%B;流速为0.4 mL·min^-1;检测波长为220 nm;动态背压(ABPR)为2000 psi;柱温为30℃。结果系统考察了色谱柱类型、动态背压(ABPR)、助溶剂种类等因素对合相色谱分离的影响,优化了最佳合相色谱条件。厚朴酚、和厚朴酚在1.5625~100 ng与峰面积呈良好的线性关系,r分别为0.9993和0.9998,平均加样回收率均≥98.4%,RSD均≤1.3%。结论该方法简便、快速、准确,可用于胃瘫外敷方中厚朴酚、和厚朴酚的含量测定,为胃瘫外敷方的质量评价和UPC2的应用提供了参考。 Objective To determine magnolol and honokiol in Weitan Waifu patch by ultra performance convergence chromatography(UPC2)and discuss the main influencing factors for its application.Methods The analysis was performed on Waters Acquity UPC2 BEH analytical column(150 mm×2.1 mm,1.7μm).The mobile phase was CO2-methanol with gradient elution.The gradient elution condition was 0-3 min:2%-10%B;3-4 min:10%-35%B;4-8 min:35%B.The flow rate was 0.4 mL·min^-1,the system back pressure was 2000 psi,the detection wavelength was 220 nm and the column temperature was 30℃.Results The effect of column type,dynamic back pressure and cosolvent type on the separation of was measured.The optimized method showed good linearity at 1.5625-100 ng for magnolol and honokiol,with a correlation coefficient(r)at 0.9993 and 0.9998,respectively.The average recoveries of magnolol and honokiol were higher than 98.4%,and the RSD was lower than 1.3%.Conclusion This method is rapid,simple,and accurate,which can be used for the determination of magnolol and honokiol in Weitan Waifu patch,which will provide evidence for the development and quality control of Weitan Waifu patch.
