摘要
目的观察神经生长因子(nerve growth factor,NGF)过表达脐带间充质干细胞(umbilical cord mesenchymal stem cells,UCMSCs)在大鼠脑出血中的治疗效果。方法建立大鼠脑出血模型并移植NGF过表达的UCMSCs细胞,观察细胞移植后大鼠神经功能、脑组织含水量和血肿体积情况。结果过表达的UCMSCs细胞移植后,NGF治疗组1 d、3 d、7 d、14 d的改良神经功能损伤评分(modified neurological severity score,mNSS)均低于脑出血组、GFP空载对照组,差异均有统计学意义(P<0.05)。与脑出血组比较,NGF治疗组移植后第1周和第2周的脑组织含水量均低于脑出血组,差异均有统计学意义(P<0.05)。与脑出血组比较,NGF治疗组移植后第1周、第2周和第4周的脑组织血肿体积均低于脑出血组,差异均有统计学意义(P<0.05)。结论NGF过表达脐带间充质干细胞治疗大鼠脑出血可改善大鼠神经功能,减少脑组织含水量及血肿体积。
Objective To evaluate the therapeutic efficacy of umbilical cord mesenchymal stem cells(UCMSCs)with over-expression of nerve growth factor(NGF)in the treatment of cerebral hemorrhage in rats.Methods Firstly,a model of cerebral hemorrhage in rats was created and UCMSCs transfected with over-expression of NGF were transplanted.Secondly,the neurological function,the water content and the volume of the hematoma of the brain tissues in the rats were monitored and documented.Results After UCMSCs transfected with over-expression of NGF were transplanted,the modified neurological severity scores(mNSS)of the NGF group as observed at 1d,3d,7d,14d were lower those of the group with cerebral hemorrhage and those in the GFP empty control group.The differences were statistically significant(P<0.05).Compared with the cerebral hemorrhage group,the water content of brain tissue in the NGF treatment group as observed at the end of the first week and that observed at the end of the second week after transplanting had been conducted were lower than those of the cerebral hemorrhage group observed at the same times.The differences were statistically significant(P<0.05).Compared with the cerebral hemorrhage group,the hematoma volume in the brain tissues in the NGF treatment group as observed 1 week,2 weeks and 4 weeks after transplantation was lower than that in the cerebral hemorrhage group as observed at the same times.The differences were statistically significant(P<0.05).Conclusions Umbilical cord mesenchymal stem cells transfected with over-expression of NGF for the treatment of cerebral hemorrhage in rats can effectively improve the neurological function of rats and reduce water content and hematoma volume in the brain tissues of rats.
