摘要
目的:探讨蒙药古日古木-13和古日古木对大鼠慢性酒精性肝纤维化是否具有保护作用及其作用机制。方法:取135只Wistar雄性大鼠,随机分为对照组,模型组,古日古木高(1.0 g·kg^-1)、中(0.5 g·kg^-1)、低(0.25 g·kg^-1)剂量组,古日古木-13高(1.0 g·kg^-1)、中(0.5 g·kg^-1)、低(0.25 g·kg^-1)剂量组,水飞蓟宾胶囊阳性组(0.04 g·kg^-1)。灌胃给药,在给药3 h后,对照组给生理盐水,其余各组均用乙醇溶液(450 mL·L^-1)灌胃,每日两次,建立慢性酒精性肝损伤模型。24周后检测大鼠血清中丙氨酸转氨酶(ALT)、天门冬氨酸转氨酶(AST)含量,取部分肝脏作HE染色和Masson染色进行病理组织学检查,检测肝匀浆中丙二醛(MDA)含量以及超氧化物歧化酶(SOD)活性。结果:模型组与古日古木-13和古日古木高剂量组相比,血清ALT、AST活力明显升高(P<0.05),肝匀浆MDA含量明显升高(P<0.05),SOD活性明显降低(P<0.05)。HE染色和Masson染色显示古日古木-13和古日古木高剂量组可有效减轻肝细胞变性、坏死和炎细胞浸润等病变及坏死浸润灶。减轻程度与古日古木-13的剂量呈正相关。结论:蒙药古日古木-13和古日古木对酒精性肝损伤具有保护作用,药物作用原理可能与其清除自由基作用、提高肝脏代谢能力、增强肝细胞膜的稳定性等有关。
Objective:To study the protective effects and mechanism of Gurigumu-13 and Gurigumu on its protection in rats of Chronic alcoholic liver fibrosis.Methods:One hundred and thirty-five wistar rats were randomly divided into nine groups:normal control group,model group,Gurigumu and Gurigumu-13 high(1 g·kg^-1),medium(0.5 g·kg^-1),low(0.25 g·kg^-1)dose groups,Silibinin capsules control group(0.04 g·kg^-1)-Model group and each dose group were fed alcohol(450 mL·L^-1)twice per day and the normal control were administered with distilled water for twenty-four weeks.There were no limited food intake during the modeling and free access to water.Take blood samples to measure ALT,AST.Take a portion of the liver for HE and Masson staining histopathological examination.Determine the MDA and SOD in the liver tissue.Results:Model group compared with the Gurigumu-13 and Gurigumu high-dose group,serum ALT,AST activity was significantly increased(P<0.05);liver tissue MDA was significantly increased(P<0.05),SOD activity decreased significantly(P<0.05).HE and Masson staining showed that Gurigumu-13 and Gurigumu could effectively reduce liver cell degeneration,necrosis,inflammatory cell infiltration and necrotic lesions infiltrates.The necrosis infiltration lesions in Gurigumu-13 and Gurigumu group significantly reduced,while the liver cell degeneration,necrosis and inflammatory cells were significantly reduced,related to the Gurigumu-13 dose.Conclusion:Studies lead us to conclude that Gurigumu-13 and Gurigumu could protect rat from chronic liver injury caused by alcoholic,and it may play an important role through its protection on liver.
