额日顿乌日勒对MCAO/R大鼠大脑皮质区BDNF及TrkBmRNA表达水平的影响 被引量:1

摘要 目的:通过观察额日顿乌日勒对大鼠脑缺血再灌注损伤后BDNF及其受体TrkBmRNA表达水平的影响,探讨治疗缺血性脑卒中的可能机制。方法:选取40只体重为180~240 g的健康雄性wistar大鼠。随机分为假手术组、模型组、治疗组(额日顿乌日勒组)、对照组(尼莫地平组)。每组10只wistar大鼠,除假手术组外其他组采用改良Zea-longa法,建立MCAO/R模型。手术造模成功后24 h开始灌胃给药,治疗组给予额日敦乌日勒500 mg/kg,对照组给予尼莫地平10 mg/kg。假手术组、模型组wistar夫鼠每31次纯净水灌胃。分别再灌注7 d后行神经功能评分、断头取脑组织。采用Real-time PCR法检测BDNF及TrkB mRNA表达的相对含量。结果:分别再灌注7 d后,与模型组相比,治疗组和对照组显著提高大鼠神经行为功能(P<0.01,P<0.05)。与假手术组相比,模型组、治疗组和对照组BDNF及TrkBmRNA表达均上调;治疗组与对照组相比BDNF及TrkBmRNA表达差异具有统计学意义(P<0.01)。结论:额日顿乌日勒能显著提高大脑中动脉阻塞再灌注局灶性脑缺血损伤模型大鼠BDNF及其受体TrkBmRNA的表达,从而达到脑缺血损伤修复和保护神经的目的。 To observe the effect of Eridun Wurile on the expression of BDNF and TrkB receptor after cerebral ischemia repeifusion injury in rats, and to explore the possible mechanism of Eridun Wurile in the treatment of ischemic stroke. Methods: Forty healthy male Wistar rats weighing 180~240 g were selected. They were randomly divided into sham-operated group, model group, treatment group (Eridun Wurile group) and control group (Nimodipine group). Ten Wistar rats in each group were used to establish MCAO/R model by modified Zea-longa method except sham operation group. 24 hours after the success of the operation, the rats were given intragastric administration with Eridun Wurile (500 mg/kg) and nimodipine (10 mg/kg). Witar rats in sham operation group and model group were given pure water once a day. After 7 days of reperfusion, neurological function score and brain tissue were obtained. Real -time PCR was used to detect the relative expression of BDNF and TrkB. Result: After reperfusion for 7 days, compared with model group, treatment group and control group could significantly improve neurobehavioral function of rats (P<0.01 ,P<0.05). Compared with the sham operation group, the expression of BDNF and TrkB in the model group and the control group were up-regulated, while the expression of BDNF and TrkB in the treatment group and the control group were up-regulated significantly (P<0.01). Conclusion: Eridun Wurile can significantly increase the expression of BDNF and TrkB receptor in rats with focal cerebral ischemia injury induced by middle cerebral artery occlusion and reperfusion. Thus, cerebral ischemia injury has the function of repairing and protecting nerves.
