摘要
目的:观察补阳还五汤对局灶性缺血模型大鼠认知能力的改善作用,同时探讨其对PI3K-AKT信号通路的影响。方法:选取30只SD大鼠按数字随机法分为假手术组、模型组及中药组,每组10只。模型组及中药组接受改良急性局灶性缺血模型制备(MCAO术)成模后中药组大鼠接受补阳还五汤灌胃,1次/d,连续干预14 d。假手术组及模型组大鼠接受等剂量的生理盐水灌胃,时间同中药组。干预结束后,比较各组行为学、脑梗死体积、海马形态学、脑组织PI3K、Akt、p-Akt蛋白/mRNA水平变化。结果:补阳还五汤可明显缩短大鼠游泳轨迹路程及逃避潜伏期,与模型组比较,差异有统计学意义(P<0.05);补阳还五汤可缩小MCAO大鼠的脑梗死体积,与模型组比较,差异有统计学意义(P<0.05);补阳还五汤可减少MCAO后大鼠海马组织病理损伤征象,上调PI3K、p-Akt、Bcl-2蛋白及mRNA表达,降低Bax蛋白及mRNA水平,与模型组比较,差异有统计学意义(P<0.05)。结论:补阳还五汤可改善MCAO大鼠认知能力,其作用机制可能与介导PI3K-AKT信号通路有关。
Objective:To observe improvement effect of Buyang Huanwu Decoction in cognitive ability of focal ischemia model rats and explore its effects on the PI3K-AKT signaling pathway.Methods:A total of 30 SD rats were randomly divided into sham operation group(n=10),model group(n=10)and Chinese materia medica group(n=10).The rats in the model group and the Chinese materia medica group received modified acute focal ischemia modeling(MCAO).After the modeling,the rats in the Chinese materia medica group received intragastric administration of Buyang Huanwu Decoction once a day for 14 d.The rats in the sham operation group and the model group were given the same dose of normal saline for the same time as the Chinese materia medica group.After the intervention,behavior,volume of cerebral infarction,morphology of hippocampus and levels of PI3K,Akt and p-Akt protein/mRNA in brain tissue were compared in each group.Results:1)Buyang Huanwu Decoction could significantly shorten the swimming track and escape latency of rats,and there was significant difference compared to that in the model group(P<0.05).2)Buyang Huanwu Decoction could reduce the cerebral infarction volume of MCAO rats,and there was significant difference compared to that in the model group(P<0.05).3)Buyang Huanwu Decoction could reduce pathological injury signs of hippocampal tissue after the MCAO,up-regulate the expressions of PI3K,p-Akt,Bcl-2 protein and mRNA,and decrease the levels of Bax protein and mRNA,which were significantly different from those in the model group(P<0.05).Conclusion:Buyang Huanwu Decoction can improve the cognitive ability of MCAO rats,and its mechanism may be related to mediating the PI3K-AKT signaling pathway.
