基于网络药理学对丹参治疗非酒精性脂肪肝病主要活性成分及潜在靶点分析 被引量:3

Analysis of components and potential targets of Salviae Miltiorrhizae(Dan Shen)in the treatment of nonalcoholic fatty liver disease based on network pharmacology

摘要 目的运用网络药理学研究方法对丹参治疗非酒精性脂肪性肝病(non-alcoholic fatty liver disease,NAFLD)主要活性成分及核心靶点进行分析。方法采用中药系统药理学分析平台(Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform,TCMSP)、DRAR-CPI分子对接平台和Uniprot数据库筛选出丹参的有效活性成分及作用靶点基因,通过Gene Cards数据库收集NAFLD疾病基因,然后对药物作用基因及疾病相关基因进行韦恩分析,确定药物作用潜在靶点,进而构建活性成分-靶点相互作用网络,并进行GO功能富集分析和KEGG通路富集分析。结果研究得到26个作用于NAFLD疾病靶点的活性成分和98个作用靶点,GO功能富集分析确定了289个条目,KEGG通路分析共发现83条作用通路。结论本研究结果初步探讨了丹参治疗NAFLD的基本药理作用及其相关机制,并为进一步的实验室研究奠定了良好的基础。 Objective To analyze the components and potential targets of Salviae Miltiorrhizae(Dan Shen)in the treatment of nonalcoholic fatty liver disease(NAFLD)based on network pharmacology.Methods Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP),Drug Repositioning and Adverse drug Reaction via Chemical-Protein Interactome(DRAR-CPI),and Universal Protein Database(Uniprot)were used to screen the active ingredients and target genes of Dan Shen.NAFLD-related genes were collected from GeneCards database,and Wayne analysis was performed on drug-acting genes and disease-related genes to determine potential targets of the drug,then active ingredient-target interaction network was constructed,and GO function enrichment analysis and KEGG pathway enrichment analysis were performed.Results A total of 26 active ingredients and 98 target sites which acted in NAFLD disease target were identified,289 items were identified by GO functional enrichment analysis,and 83 pathways were identified by KEGG pathway analysis.Conclusion This study preliminarily discusses the basic pharmacological effect and related mechanism of Dan Shen on NAFLD,which has laid a good foundation for further laboratory research.
