摘要
目的分析线粒体分布和形态调节因子1(MSTO1)在多种肿瘤中的表达、预后及作用机制。方法利用癌症基因组图谱计划(TCGA)门户网站UALCAN泛癌症分析MSTO1的表达变化及其与预后的关系;用LinkedOmics网站和DAVID 6.8网站进行基因本体(GO)和KEGG信号通路分析。结果在膀胱尿路上皮癌、结肠癌、肝脏肝细胞癌、肺腺癌、直肠腺癌和胃腺癌中MSTO1基因表达较各自正常组织显著上调(P<0.01);MSTO1高表达利于卵巢癌和肾上腺皮质瘤预后(P<0.05);MSTO1高表达不利于肾透明细胞瘤、肝脏肝细胞癌、脑低级神经胶质瘤和黑色素瘤的预后(P<0.05)。基因本体(GO)和KEGG信号通路分析发现MSTO1共表达蛋白主要定位于细胞质和细胞核等,主要参与细胞分裂及细胞周期调控等过程,功能包括结合蛋白质、多聚腺苷酸RNA、ATP、核糖体等。结论MSTO1有可能成为预测肿瘤预后的潜在标志物,为肿瘤生物学研究提供了一个新的思路。
Objective To investigate the expression and prognostic significance and functional mechanism of MSTO1(mitochondrial distribution and morphogenetic factor 1)by pan-cancer analysis.Methods We analyzed?the expression of MSTO1 and its relationship with the prognosis of some types of cancer by using the UALCAN and Pan-Cancer Project(TCGA)portal website.And the gene ontology(GO)and KEGG signaling pathway were analyzed by using the Linked Omics website and DAVID 6.8 website.Results MSTO1 expressed higher significantly in bladder urothelial carcinoma,colon cancer,liver cancer,lung adenocarcinoma,rectal adenocarcinoma and gastric adenocarcinoma than in normal tissues(P<0.001,P<0.01).The higher expression ofMSTO1 was helpful to the prognosis of ovarian cancer(P=0.027)and adrenal cortical tumor(P<0.001).Higher expression ofMSTO1 was unfavorable to the prognosis of renal clear cell tumor,liver cancer,low-grade glioma,melanoma(P<0.05).The KEGG showed that MSTO1 coexpressed proteins mainly located in the cytoplasm and nucleus,involved in cell division,cell cycle regulation,and other processes.Results from GO showed that functions of MSTO1 coexpressed proteins generally included binding proteins,polyadenylate RNA,ATP,ribosome,etc.Conclusion MSTO1 may be a potential marker for predicting the prognosis of a tumor,which provides a newidea for tumor biology research.
