摘要 目的探究神经菌毛素-1(NRP-1)靶向肽EG3287对人口腔鳞状细胞癌化疗敏感性的影响。方法首先构建NRP-1敲低的Tca8113细胞株(KD-Tca8113)和BcaCD885细胞株(KD-BcaCD885),采用CCK-8试剂盒检测野生型和NRP-1敲低型细胞株对化疗顺铂的敏感性,计算半数致死剂量(IC50)。以乱序肽为对照,评估NRP-1靶向肽EG3287对野生型Tca8113和BcaCD885细胞对顺铂敏感性的影响。结果流式细胞术结果显示,EG3287肽对NRP-1具有明显的靶向作用;CCK-8结果显示,顺铂对WT-Tca8113、KD-Tca8113、WT-BcaCD885和KD-BcaCD885的IC50分别为(5.32±0.44)、(0.56±0.02)、(4.98±0.57)和(0.64±0.03)μg/ml。野生型细胞IC(50)明显比NRP-1敲低细胞高(P<0.01)。在IC(50)剂量的顺铂处理下,EG3287肽组的细胞凋亡率明显较空白对照组和乱序肽组高(P<0.01)。结论NRP-1表达量与人口腔鳞状细胞癌Tca8113和BcaCD885细胞株对顺铂的敏感性呈负相关,NRP-1靶向肽能明显提高细胞对顺铂的敏感性。 Objective To explore the effect of neuropilin-1(NRP-1)-targeting peptide EG3287 on sensitivity of human oral squamous cell carcinoma cell line to chemotherapy.Methods First,the cell line Tca8113(KD-Tca8113)and cell line BcaCD885(KD-BcaCD885)knocked down by NRP-1 were constructed.Then,Cell Counting Kit-8(CCK-8)was used to detect the sensitivity of wild-type cell line and NRP-1 knockdown-type cell line to the chemotherapy drug cisplatin,and the half maximal inhibitory concentration(IC50)was calculated.The effect of NRP-1-targeting peptide EG3287 on the sensitivity of wild-type Tca8113 and BcaCD885 cells to cisplatin was evaluated using scrambled peptides as controls.Results The results of flow cytometry showed that peptide EG3287 had obvious targeting effect on NRP-1.The results of CCK-8 assay showed that the IC50 of cisplatin for WT-Tca8113,KD-Tca8113,WT-BcaCD885 and KD-BcaCD885 were(5.32±0.44)μg/ml,(0.56±0.02)μg/ml,(4.98±0.57)μg/ml and(0.64±0.03)μg/ml,respectively.The IC50 of wild-type cells was significantly higher than that of NRP-1 knockdown-type cells(P<0.01).After the treatment of cisplatin(dose:IC50),the apoptosis rate of EG3287 peptide group was significantly higher than that of the blank control group and the scrambled peptide group(P<0.01).Conclusion The expression of NRP-1 is negatively correlated with the sensitivity of human oral squamous cell carcinoma cell lines Tca8113 and BcaCD885 to cisplatin.The NRP-1-targeting peptide can significantly increase the sensitivity of cells to cisplatin.
