乌头属5种植物ITS2和psbA-trnH鉴别适用性研究 被引量:2

摘要 【目的】基于传统分类鉴定,对乌头属5种植物显柱乌头系的长喙乌头Aconitum georgei H. F. Comber和石膏山乌头Aconitum rockii var. fengii(W. T. Wang)W. T. Wang、蔓乌头系的瓜叶乌头Aconitum hemsleyanum E. Pritzel和玉龙乌头Aconitum stapfianum Handel-Mazzetti、兴安乌头系的中甸乌头Aconitum piepunense Handel-Mazzetti的ITS2区和psbA-trnH的鉴别适用性进行考察。【方法】以ITS2F/ITS2R和psbA/trnH为引物,对5种乌头进行PCR扩增、ITS2和psbA-trnH的序列特征分析、系统发育树构建。【结果】ITS2位点可准确鉴定显柱乌头系的长喙乌头和石膏山乌头以及蔓乌头系的玉龙乌头,而不适用于鉴别蔓乌头系的瓜叶乌头和兴安乌头系的中甸乌头;psbA-trnH位点不适用于本研究中的5种乌头鉴别。【结论】ITS2和psbA-trnH鉴别乌头属植物均具有一定的局限性,二者对乌头属植物的鉴别适用性不同。 Objective Based on traditional classification and identification,to investigate the applicability of ITS2 and psbA-trnH for identifying 5 species of Aconitum including Aconitum georgei H. F. Comber and Aconitum rockii var. fengii(W. T. Wang) W. T. Wang in Ser. Stylosa,Aconitum hemsleyanum E. Pritzel and Aconitum stapfianum Handel-Mazzetti in Ser. Volubilia,and Aconitum piepunense Handel-Mazzetti in Ser. Ambigua. Methods With ITS2F/ITS2R and psbA/trnH as primers, polymerase chain reaction(PCR) amplication was carried out for analyzing the sequence characteristics and constructing the phylogenetic tree for 5 species of Aconitum. Results ITS2 was suitable for identification of A. georgei and A. rockii var. fengii in Ser. Stylosa,and A. stapfianum in Ser.Volubilia instead of A. hemsleyanum in Ser. Volubilia and A. piepunense Ser. Ambigua. PsbA-trnH was not suitable to identify 5 species of Aconitum. Conclusion ITS2 and psbA-trnH have different applicabilities for Aconitum species,but have certain limitation.
