电针对脑卒中肢体痉挛大鼠皮质BDNF、TrkB及GABAa表达的影响 被引量:19

Effects of Electroacupuncture on Expressions of BDNF,TrkB and GABAa in Cortex of Rats with Limb Spasm after Stroke

摘要 目的观察电针对脑卒中肢体痉挛大鼠皮质脑源性神经营养因子(BDNF)、酪氨酸激酶受体B(TrkB)、γ-氨基丁酸(GABA)受体(GABAa)表达的影响,探讨电针治疗脑卒中肢体痉挛的机制。方法将77只SD雄性大鼠随机分为空白组、假手术组各9只及模型储备组59只,运用Zea Longa线栓结合内囊注射NMDA受体制作脑卒中肢体痉挛大鼠模型。18只成模大鼠随机分为模型组、电针组。电针组取双侧阳陵泉、曲池针刺,每次30 min,每日1次,连续5 d;假手术组和模型组同期只固定不作任何干预,空白组不作任何处理。观察各组大鼠Zea Longa评分,改良Ashworth肌张力量表评分,皮质BDNF、TrkB、GABAa mRNA及蛋白表达。结果与空白组、假手术组比较,模型组和电针组大鼠治疗前行为学评分明显升高(P<0.01);与模型组比较,电针组大鼠治疗后行为学评分明显降低(P<0.05);与空白组、假手术组比较,模型组大鼠皮质BDNF、TrkB、GABAa mRNA及蛋白表达明显降低(P<0.05);与模型组比较,电针组大鼠皮质BDNF、TrkB、GABAa mRNA及蛋白表达明显升高(P<0.05)。结论电针可改善脑卒中肢体痉挛大鼠神经功能评分及肌张力,其机制可能与调节模型大鼠皮质BDNF、TrkB、GABAa表达相关。 Objective To observe the effects of electroacupuncture on the expressions of BDNF,TrkB and GABAa in the cortex of rats with limb spasm after stroke;To explore the mechanism of electroacupuncture in the treatment of limb spasm after stroke.Methods Totally 77 SD male rats were randomly divided into blank group(9 cases),sham-operation group(9 cases),and model reserve group(59 cases).Rat model with limb spasm after stroke was prepared by using Zea Longa suture-occluded and injecting of NMDA receptor in inner capsule.Then,the successfully prepared 18 rats were randomly divided into the model group and the electroacupuncture group.In the electroacupuncture group,electroacupuncture was used to stimulate Yanglingquan(GB34)and Quchi(LI11)once a day for 30 min each time,for consecutive 5 d.The sham-operation group and the model group just fixed at the same time without any intervention,while the blank group received no treatment.Zea Longa score,the modified Ashworth Tension Scale,and the mRNA and protein expressions of BDNF,TrkB,and GABAa in the cortex were observed.Results Compared with the blank group and the sham-operation group,the behavioral score of model group and electroacupuncture group increased significantly(P<0.01).After treatment,compared with the model group,the behavioral score of the electroacupuncture group decreased significantly(P<0.05).Compared with the blank group and the sham-operation group,the mRNA and protein expressions of BDNF,TrkB,and GABAa in the cortex of the model group decreased significantly(P<0.05),while the mRNA and protein expressions of BDNF,TrkB,GABAa in the cortex of the electroacupuncture group increased significantly compared with the model group(P<0.05).Conclusion Electroacupuncture can improve neurological function score and muscle tone in rats with limb spasm after stroke,which may be through the regulation of cortical BDNF,TrkB,and GABAa.
