硫酸亚铁法和Fontana法显示黑色素的比较研究 被引量:1

摘要 目的:比较硫酸亚铁法与Fontana法对皮肤黑色素颗粒的染色效果。方法:用中波红斑效应紫外线(UVB)照射豚鼠皮肤以制造动物模型。利用硫酸亚铁法和Fontana法对正常豚鼠皮肤和照射UVB的豚鼠皮肤进行黑色素染色,对比两种不同黑色素染色方法的效果。结果:硫酸亚铁法与Fontana法均能对豚鼠皮肤黑色素进行着色,显示照射UVB的豚鼠皮肤含黑色素颗粒的细胞数比正常豚鼠皮肤的明显增多。硫酸亚铁法显示黑色素颗粒为墨绿色,Fontana法显示黑色素颗粒为黑色,但Fontana法也使亲银颗粒和嗜铬颗粒显示为黑色。结论:硫酸亚铁法检测黑色素与Fontana法相比,染色特异性高,更适合于临床病理检查及实验室研究。 Objective:To compare the staining effects of ferrous sulfate method and Fontana method on melanin granules.Methods:The skin of guinea pig was irradiated with UVB to make animal model.The melanin staining of normal guinea pig skin and UVB irradiated guinea pig skin was carried out by ferrous sulfate method and Fontana method.The results of two different methods of melanin staining were compared.Results:Both ferrous sulfate method and Fontana method can stain the melanin in the skin of guinea pigs,which shows that the number of melanocytes in the skin of guinea pigs exposed to UVB is significantly higher than that of normal guinea pigs.Ferrous sulfate method showed that the melanin particles were dark green,Fontana method showed that the melanin particles were black,but Fontana method also showed that the argentaffin particles and chromaffin particles were black.Conclusion:Compared with Fontana method,ferrous sulfate method is more specific in detecting melanin,which is more suitable for clinical pathological examination and laboratory research.
