摘要
目的观察肾炎康复片对糖尿病大鼠肾组织骨形态蛋白-7(BMP-7)、转化生长因子β1(TGF-β1)及Smad1表达的影响.方法将SD大鼠随机分为正常对照组(NC组)、糖尿病模型组(DM组)和肾炎康复片干预组(SYK组).用链脲佐菌素(STZ)诱导大鼠糖尿病模型.SYK组以肾炎康复片600mg·kg^-1·d^-1的剂量灌胃给药,其余两组大鼠给予相当剂量生理盐水灌胃.8周末检测各组大鼠血糖、血肌酐(Scr)、尿素氮(BUN)、24h尿蛋白(Upro),计算肾小球平均体积(MGV),应用免疫组化法检测肾小球BMP-7、TGF-β1和Smad1的表达.结果DM组Scr、BUN、Upro和MGV较NC组明显升高(P<0.05),SYK组上述各指标明显低于DM组(P<0.05).免疫组化结果显示,与NC组比较,DM组肾小球BMP-7表达降低,TGF-β1、Smad1表达升高(P<0.05);SYK组BMP-7表达高于DM组(P<0.05),TGF-β1、Smad1表达低于DM组(P<0.05).结论肾炎康复片对糖尿病大鼠肾脏的保护作用可能与降低TGF-β1、Smad1表达和升高BMP-7表达有关.
Objective To investigate the effect of Shenyankangfu tablet on the expression of bone morphogenetic protein-7(BMP-7),transforming growth factor-β1(TGF-β1)and Smad1 in the kidney of diabetic rats.Methods SD rats were randomly divided into normal control group(NC group),diabetic model group(DM group)and Shenyankangfu tablet group(SYK group).The diabetic rat model was induced with streptozotocin.Rats in SYK group were given Shenyankangfu tablet(600mg·kg-1·d-1)by gavage once a day.Rats in the other two groups were given the same dose of saline by gavage.After 8 weeks,blood glucose,serum creatinine(Scr),blood urea nitrogen(BUN)and 24-hour urinary protein(Upro)were measured.Mean glomerular volume(MGV)was calculated.The expression of BMP-7,TGF-β1 and Smad1 were detected by immunohistochemistry.Results The Scr,BUN,Upro and MGV were significantly higher in DM group than those in NC group(P<0.05),and those in SYK group decreased compared with DM group(P<0.05).Compared with NC group,the expression of BMP-7 was lower and the expression of TGF-β1 and Smad1 were higher in DM group(P<0.05).Rats in SYK group had higher expression of BMP-7 and lower expression of TGF-β1 and Smad1 compared with DM group(P<0.05).Conclusion Shenyankangfu tablet can protect the kidney of diabetic rats,which may mediate through increasing the expression of BMP-7 and decreasing the expression of TGF-β1 and Smad1.
