布鲁氏杆菌S2株DUF2326基因克隆与生物信息学分析 被引量:1

摘要 为了分析布鲁氏杆菌猪种S2疫苗株DUF2326基因编码蛋白的结构、功能以及抗原表位,试验对DUF2326基因进行克隆、生物信息学分析及抗原表位预测。结果表明:DUF2326基因全长1653 bp,编码550个氨基酸,分子质量为61.775 ku,理论等电点为5.34,呈酸性,不稳定指数为35.17,该蛋白无跨膜区,不存在信号肽,α-螺旋为二级结构的主要成分(59.64%)。DUF2326蛋白是一种抗原性较高的亲水性蛋白,含有较多潜在的B细胞抗原表位。说明DUF2326蛋白具有良好的抗原性,具备成为鉴别布鲁氏杆菌病疫苗免疫和自然感染抗原的潜力。 In order to analyze the structure,function and antigenic epitope of the DUF2326 protein encoded by the DUF2326 gene of Brucella vaccine strain S2,this experiment cloned,bioinformatics and epitope prediction of DUF2326 gene.The results showed that DUF2326 gene was 1653 bp in full length.It encoded a protein of 550 amino acids.The protein weighed 61.775 ku.The theoretical pI was 5.34,The instability index was 35.17.The protein has no transmembrane region and no signal peptide,α-Helix is the main component of the secondary structure(59.64%).DUF2326 protein was a hydrophilic protein with high antigenicity,It contained more potential B cell epitopes.This indicates that DUF2326 protein has good antigenicity and has the potential to be an antigen for identifying vaccine immunity and natural infection of brucellosis.
