摘要
为了探究濒危物种黑颈鹤(Grus nigricollis)的肠道真菌群落多样性和优势菌群,试验采用从草海收集的3份新鲜的黑颈鹤粪便进行真菌多样性初探,利用高通量测序技术中的Illumina HiSeq平台对转录间隔区1(ITS1)区域进行测序,对操作分类单元(OTUs)进行聚类分析及α、β多样性指数分析。结果表明:从3份样品中共获得221048条Tags,平均构建OTUs为367个,共有OTUs仅为7个,主要包括4个门、8个纲、10个目、9个科和5个属。其中子囊菌门(Ascomycota,平均含量为46.9%)和担子菌门(Basidiomycota,平均含量为45.8%)为优势门,前者以座囊菌纲(Dothideomycetes)、锤舌菌纲(Leotiomycetes)和枝孢菌属(Cladosporium)、微白粉菌属(Microidium)为主,后者以伞菌纲(Agaricomycetes)、微球黑粉菌纲(Microbotryomycetes)和散尾鬼笔属(Lysurus)、红酵母属(Rhodotorula)为主。α和β多样性指数分析均说明黑颈鹤肠道菌群的种类和数量的丰富度极高,多样性差异显著。
In order to explore the diversity and dominant flora of intestinal fungi community of endangered Grus nigricollis,transcriptional interval region 1(ITS1)of three fresh feces collected from Caohai were sequenced using the Illumina HiSeq platform in high-throughput sequencing technology.The operational taxonomic units(OTUs)was performed cluster analysis and diversity index analysis ofαandβ.The results showed that a total of 221,048 Tags have been obtained from the three samples,and the average number of Tags constructed is 367,only 7 was owned by the three,which mainly includes 4 phyla,8 classes,10 orders,9 families and 5 genera.Ascomycota(46.9%)and Basidiomycota(45.8%)were the dominant phylum.The former was dominated by Dothideomycetes,Leotiomycetes,Cladosporium and Microidium spp.The latter were dominated by Agaricomycetes,Microbotryomycetes,Lysurus and Rhodotorula.The analysis ofαandβdiversity index indicated that the intestinal flora of black-neck-crane were extremely rich in species and number,and the diversity difference was significant.
