猪细环病毒Ⅰ型实时荧光定量PCR检测方法的建立

摘要 为了建立一种能在临床上快速、准确检测猪细环病毒I型(Torque teno sus virus 1,TTSuV1)的实时荧光定量PCR方法,试验根据GenBank上公布的TTSuV1序列中的高度保守片段设计引物及探针,并构建标准质粒;测定标准质粒浓度,以标准质粒为模板建立检测TTSuV1的TaqMan实时荧光定量PCR方法,并对该方法的灵敏度、稳定性和特异性进行评价。结果表明:构建的标准质粒浓度为3.3×1011copeis/μL;建立的实时荧光定量PCR最佳退火温度为54.5℃,最佳引物浓度为0.8 mol/μL,探针浓度为0.4 mol/μL;所建立的实时荧光定量PCR检测方法具有良好的灵敏性、稳定性和特异性。说明该检测方法可应用于临床检测TTSuV1。
