摘要
目的:探讨精液冷冻前优化处理方式对不同精子活力的精子冷冻复苏效果的影响。方法:选取符合条件的标本50份,每份标本等分4份,以原液作为对照(A组),分别采用三种不同方式[洗涤法(B组)、上游法(C组)和密度梯度离心法(D组)]进行处理,再行精子冷冻复苏。比较不同精子活力标本在不同优化方法处理后的精子冷冻复苏率。结果:精子冷冻复苏率由高至低排序为A组>C组>D组>B组。B组和D组精子冷冻复苏率均显著低于A组(P<0.05),C组与A组比较差异无统计学意义(P>0.05)。低活力(前向运动精子PR≤32%)标本中,各处理方式精子冷冻复苏率比较差异无统计学意义(P>0.05)。正常活力(前向运动精子PR>32%)标本中,B组精子冷冻复苏率显著低于其余三组(P<0.05),A组、C组、D组三组比较差异无统计学意义(P>0.05)。结论:精液冷冻前优化处理方式对精子冷冻复苏率有影响,实际工作中,应根据精液的质量选择合适的精液冷冻前优化处理方式。
Objective:To investigate the effect of spermatozoa selection before freezing on the recovery rate of human frozen spermatozoa of different sperm motility.Method:A total of 50 specimens that meet the requirements were selected.Each sample was divided into 4 equal parts,and the stock solution was used as a control (group A).Before sperm freezing and recovery,the spermatozoa selection was carried out in three different ways [washing (group B),swimming-up (group C) and density-gradient centrifugation(group D)].The recovery rate of frozen spermatozoa of different sperm motility with different treatment was compared.Result:The sperm recovery rate was ranked from high to low in group A > group C > group D > group B.The sperm recovery rate of group B and group D were significantly lower than those of group A (P<0.05),and there was no significant difference between group C and group A (P>0.05).In low vitality (PR≤32%),there was no significant difference in the sperm recovery rates between the treatments (P>0.05).In normal vitality (PR>32%),the sperm recovery rate in group B was significantly lower than those of the other three groups (P<0.05),and there was no significant difference among group A,group C,and group D (P>0.05).Conclusion:The spermatozoa selection before freezing has an effect on the recovery rate of frozen spermatozoa.In actual work,the appropriate semen pre-freezing treatment should be selected according to the quality of semen.
