RP-HPLC法同时测定莲子心提取物中4种生物碱的含量

Simultaneous determination of 4 alkaloids in Lotus plumule extract by RP-HPLC

目的应用反相高效液相色谱法(RP-HPLC)同时测定莲子心提取物中莲心碱、异莲心碱、甲基莲心碱和莲心季铵碱4种生物碱的含量。方法色柱谱:ZORBAX SB-C 18(150 mm×4.6 mm,5μm);流动相:乙腈-水-三乙胺(磷酸调节pH值为3.3);流速:0.8 mL·min^-1;检测波长:282 nm;柱温:30℃;进样量:20μL。结果莲心碱、异莲心碱、甲基莲心碱和莲心季铵碱分别在0.4~4.9,0.5~6.1,0.0525~0.6200和0.614~2.158μg·mL-1范围内呈良好的线性关系(r>0.9994),且平均回收率较好(RSD<2.3%);样品中4种生物碱的含量分别为66.95,38.71,62.43和35.82μg·mL-1。结论RP-HPLC法可用于同时测定莲子心提取物中莲心碱、异莲心碱、甲基莲心碱和莲心季铵碱的含量,该方法简便、高效,灵敏度高,具有较好的重复性,可为莲子心提取物的质量控制提供参考。

Objective To determine the contents of 4 alkaloids of liensinine,isoliensinine,neferine and lotusine in Lotus plumule extract simultaneously by reversed-phase high-performance liquid chromatography(RP-HPLC).Methods Chromatography:ZORBAX SB-C 18(150 mm×4.6 mm,5μm);mobile phase:acetonitrile-water-triethylamine(pH 3.3 adjucted by phosphoric acid);flow rate:0.8 mL·min-1;detection wavelength:282 nm;column temperature:30℃;injection:20μL.Results The linear relationships of liensinine,isoliensinine,neferine and lotusine in the range of 0.4-4.9,0.5-6.1,0.0525-0.6200 and 0.614-2.158μg·mL^-1 were good(r>0.9994),and the sample recovery rate was satisfactory(RSD<2.3%);the contents of 4 alkaloids were 66.95,38.71,62.43 and 35.82μg·mL-1 respectively.Conclusion RP-HPLC can be used for the simultaneous determination of liensinine,isoliensinine,neferine and lotusine,the method is simple,efficient,with high sensitive and good repeatability.This study provides an effective reference for the quality control of Lotus plumule extract.
