圆叶节节菜不同体积分数乙醇提取物的止血和肝保护作用研究 被引量:1

摘要 目的研究提取溶剂中不同体积分数乙醇对圆叶节节菜止血作用的影响。方法采用毛细管法、半自动凝血分析仪和全自动三分群血液分析仪测定凝血时间(CT)、凝血酶原时间(PT)、活化部分凝血活酶时间(APTT)、血浆凝血酶时间(TT)、血小板数(PLT)和血小板分布宽度(PDW),评价圆叶节节菜不同体积分数(5%,10%,20%,50%,80%和100%)乙醇提取物体内外止血作用;采用四氯化碳(CCl 4)诱导小鼠急性肝损伤模型,测定小鼠血清谷丙转氨酶(ALT)、总蛋白(TP)和总胆红素(TB)的水平;以具有最佳止血效果的圆叶节节菜提取物作为研究对象,采用PT、APTT和TT测定法研究量效关系。结果随着乙醇体积分数的提高,圆叶节节菜乙醇提取物产率下降;乙醇体积分数为50%及以上的圆叶节节菜提取物中CT、PT、APTT和TT显著缩短;与对照组比较,乙醇体积分数为50%,80%和100%的提取物中PLT显著增加,但各治疗组的PDW差别不显著;各提取物组的TP水平变化不大,与对照组比较,乙醇体积分数为5%,10%和100%提取物组的TB水平显著下降,乙醇体积分数高于20%的各提取物组ALT水平显著降低;乙醇体积分数为100%提取物组止血作用最佳,且具有剂量依赖性。结论提取溶剂中乙醇的体积分数对圆叶节节菜止血作用有重要影响,高体积分数乙醇提取物具有显著的止血作用。 Objective To study the effect of ethanol different volume fraction in extraction solvent on hemostasis of Rotala rotundifolia.Methods Capillary tube method,semi-automatic coagulation analyzer and automatic three-clustering blood analyzer were used to measure the coagulation time(CT),prothrombin time(PT),activated partial thromboplastin time(APTT),plasma thrombin time(TT),platelet count(PLT)and platelet distribution width(PDW)for evaluating the hemostatic effect of different volume fractions(5%,10%,20%,50%,80%and 100%)of ethanol extracts from Rotala rotundifolia in vivo and in vitro.Carbon tetrachloride(CCl 4)was used to induce acute liver injury in mice,and serum alanine aminotransferase(ALT),total protein(TP)and total bilirubin(TB)in mice were measured.By taking the Rotala rotundifolia extract with the best hemostatic effect as the research object,the dose-effect relationship was studied by PT,APTT and TT methods.Results With the increase of ethanol volume fraction in extracting solvent,the yield of Rotala rotundifolia extract decreased;Rotala rotundifolia extracts with ethanol volume fraction of 50%or more significantly shortened the CT,PT,APTT and TT.Compared with the control group,PLT of 50%,80%and 100%ethanol extract increased significantly,but the difference of PDW of each treatment group was not significant;the TP levels of each extract group changed little.Compared with the control group,the TB levels of 5%,10%and 100%ethanol extract group decreased significantly,and the ALT level of 20%ethanol extract and above extract group decreased significantly.Hemostasis of 100%ethanol extract was the best and dose-dependent.Conclusion The volume fraction of ethanol in extraction solvent has an important influence on the hemostasis of Rotala rotundifolia,and the high volume fraction ethanol extract has a significant hemostasis effect.
