摘要
目的:建立可见-紫外分光光度法检测丹眠颗粒总皂苷的含量。方法:采用岛津UV-2501PC可见-紫外分光光度计,检测波长为560 nm。以人参皂苷Re为对照品,经显色测定吸光度,绘制标准曲线计算含量。结果:总皂苷在0.04~0.20μg/mL范围内线性关系良好,回归方程为Y=3.6673 x-0.0222,r=0.9990;精密度、重复性、稳定性试验RSD均小于2.0%;平均回收率为98.46%(RSD为1.56%,N=6)。结论:本实验方法简便准确、重复性好、灵敏度高、含量测定方法稳定可行,适用于丹眠颗粒中总皂苷的含量测定,可更好控制其质量。
Objective:The UV spectrophotometry was established to detect the total saponin content of Danmian granules.Methods:Using Shimadzu UV-2501PC visible-ultraviolet spectrophotometer,the detection wavelength is 560 nm.Ginseng saponins Re was used as reference substance to determine absorbance by color rendering standard curve to calculate content.Results:A good linear relationship of total saponin was in the range of 0.04~0.20μg/mL.The regression equation was Y=3.6673x-0.0222,r=0.9990.The RSD values of precision,repeatability and stability test were less than 2.0%.The average recovery was 98.46%(RSD is 1.56%,N=6).Conclusion:The method is simple,accurate,reproducible,sensitive,and the method is stable and feasible.It is suitable for determination the content of total saponin in Danmian granules,which is better for control its quality.
