摘要
目的探讨白及二氢菲类化合物对临床常见病原菌的作用,为白及药理活性成分的临床应用提供实验依据。方法实验以琼脂稀释法测定不同白及组织的二氢菲类化合物(Dihydrophenanthropyrans,Dihs)对金黄色葡萄球菌等6种常见病原菌及标准菌株的体外最低抑菌浓度(MIC),常见病原菌均采自同一科室的临床菌株,实验时间从2017年5月至2018年5月。接着将菌株接种到Dihs浓度为MIC的培养基及对照培养基上,共培养48 h进行抗生素药敏试验,然后测量抑菌圈直径,结合菌斑图,判断Dihs对临床菌株及抗生素的药理作用。结果白及Dihs具有抑制临床病原菌生长的作用,且不同材料的Dihs对菌株的MIC值影响不尽相同,其中悬浮细胞组织中的Dihs对金黄色葡萄球菌的MIC值相对于其他白及组织最小,为(0.58±0.23)mg/mL。同时,Dihs有增强病原菌对抗生素敏感性的作用,但是对某些抗生素Dihs也有降低病原菌对抗生素的敏感性的作用,其中,Dihs增强白假丝酵母菌对抗生素敏感性作用较大,抑菌圈直径平均增加了(4.4±0.3)mm。进一步从实验结果还可以发现,白及Dihs具有逆转某些病原菌耐药性的作用。结论白及二氢菲类化合物对临床常见病原菌具有抑制作用和逆转耐药性的作用。本研究为白及的药理学研究提供了基础数据的参考,也为江油地区白及的抑菌等活性成分的利用和临床新型抗生素的研究提供理论依据。
Objective To investigate the effects of dihydrophenanthropyrans of Bletilla striata on common pathogenic bacteria,and to provide experimental evidence for the clinical application of Bletill astriata’s pharmacological active ingredients.Methods In vi tro minimum inhibitory concentration(MIC)of dihydrophenanthropyrans(Dihs)from different tissues of Bletilla striata against 6 common pathogens including staphylococcus aureus and standard strains was determined by AGAR dilution method.The common pathogens were all clinical strains from the same department,and the experimental time was from May 2017 to May 2018.Then the strain was inoculated into the medium with MIC of Dihs concentration and the control medium,and incubated for 48 h for antibiot ic susceptibility test.Then,the diameter of the inhibition zone was measured and the pharmacological effects of Dihs on clinical strains and antibiotics were determined according to the plaque diagram.Results The results showed that Dihs of Bletilla striata could inhibit the growth of clinical pathogenic bacteria,and the MIC values of Dihs in different tissues had different effects on bac terial strains.Compared with other tissues of Bletilla striata,the MIC values of Dihs in suspended cell tissue to Staphylococcus aure us was the lowest,which was 0.58±0.23 mg/mL.Meanwhile,Dihs could enhance the sensitivity of pathogenic bacteria to antibiotics,but Dihs could also reduce the sensitivity of pathogenic bacteria to some antibiotics.Among them,Dihs enhanced the sensitivity of Candida albicans to antibiotics,and the diameter of inhibition circle increased by 4.4±0.3 mm on average.Furthermore,it was found that Dihs of Bletilla striata could reverse the drug resistance of some pathogens.Conclusions Dihs of Bletilla striata could inhibit the growth of clinical pathogenic bacteria,and reverse the drug resistance of some bacteria.This study provides basic data for the pharmacological research of Bletilla striata and theoretical basis for the utilization of antibacterial and other active ingredients of Bletilla striata in Jiangyou area and the research on new clinical antibiotics.
