摘要
香豆酸-3-羟化酶(p-coumarate 3-hydroxylase, C3H)与木质素生物合成途径密切相关,是细胞色素P450中的一员。本研究利用生物信息学方法,对29条辣椒C3H蛋白序列进行分析,预测其一级结构理化特性、基序、二级结构和三级结构,并用MEGA 5.0在线软件把拟南芥与辣椒的C3H蛋白序列进行比对分析,构建系统进化树。结果表明:辣椒的C3H蛋白长度介于152~983 aa,分子量和等电点分别介于16 926.8~111 899和4.600 8~9.370 6之间。基序预测发现辣椒C3H家族的10个保守基序中最长的含50个氨基酸;α-螺旋和无规则卷曲是主要的二级结构,三级结构中都含有螺旋-转角-螺旋结构。进化树分析表明CA00g62090与AT5G26749.1同源性高,CA09g17320与AT3G51120.1同源性高,推测辣椒的CA00-g62090与CA09g17320可能较高程度地参与了木质素合成代谢调控。这为下一步研究C3H基因功能提供科学数据。
P-coumarate 3-hydroxylase(C3H)belongs to the CYP98 subfamily of cytochrome P450 and is one of the key enzymes in the lignin biosynthesis pathway.In this study,the bioinformatics method was used to analyze the sequence of 29 C3H protein samples,to predict the physicochemical properties,motif,secondary structure and tertiary structure of molecular weight and isoelectric point.With MEGA 5.0,63 C3H of Arabidopsis thaliana and 29 C3H of Capsicum were constructed to a phylogenetic tree.The results showed that the protein length of C3H in Capsicum was 152~983 aa,and the molecular weight and isoelectric point were between 16926.8~111899 and 4.6008~9.3706 respectively.There are 10 conserved motifs of the Capsicum C3H,and the longest base sequence contains 50 amino acids;α-helix and random coil are the main secondary structures,and the tertiary structure contains the helix-turn-helix structure.In the phylogenetic tree analysis,CA02g23810 has high homology with AT1G66810.1,so dose the CA11g20080 and AT3 G19360.1.It is speculated that CA02g23810 and CA11g20080 of Capsicum may be involved in the regulation of lignin anabolism,which provides a theoretical reference for further study of C3H gene function.
